ObjectiveTo investigate the immunoregulatory effect of Purple Sweet Potato Anthocyanins(PSPA),spleen lymphocytes from the mice were taken as target cells in vitro.The immune function model was established for studying the immunomodulatory activity of PSPA.This study would provide experimental basis for the development and utilization of PSPA.MethodsThe immune function model of spleen lymphocytes from the mice was established for studying the immunomodulatory activity of PSPA.The effect of cell proliferation was detected by MTT method in the different concentration PSPA groups,PSPA+concanavalin A(Con A)group,PSPA + lipopolysaccharide(LPS)and PSPA + lymphopoiesis infected by staphylococcus aureus groups.The effect of spleen lymphocytes secrete cytokines was detected by ELISA kits in the different concentration PSPA groups and PSPA + lymphopoiesis infected by staphylococcus aureus groups.The effect of expression level of cytokine gene of IL-2,IL-10,IL-13 and TNF-? were detected by RT-PCR method in the different concentration PSPA groups and PSPA + lymphopoiesis infected by staphylococcus aureus groups.ResultsThe stimulation index(SI)of lymphocyte was increased in the different concentration PSPA groups,PSPA + Con A group,PSPA + LPS group andPSPA + lymphopoiesis infected by staphylococcus aureus groups.SI of PSPA+ Con A group and PSPA + LPS group was higher than that of the different concentration PSPA groups.When lymphocytes were infected by Staphylococcus aureus,SI of the different concentration PSPA groups were increased,but,these were lower than that of normal group.The effects of IL-2,IL-10,IL-13 and TNF-? lymphocytes secretion were increased in the different concentration PSPA groups.When lymphocytes were infected by Staphylococcus aureus,the effects of IL-2,IL-10,IL-13 and TNF-? lymphocytes secretion in the different concentration PSPA groups was lower than that of nomal group.The gene expression levels of IL-2,IL-10,IL-13 and TNF-? m RNA could be up-regulated in the different concentration PSPA groups.ConclusionsIn concentration range of 2.5 mg/m L~20 mg/m L,PSPA could single s timulate the proliferation of spleen lymphocyte and conjunction with Con A or LPS.When lymphocytes were infected by Staphylococcus aureus,The effects of the different concentration PSPA groups to lymphocyte prolifera tion ability were lower than normal group.The different concentration PSPA could induce the secretion of IL-2,I L-10,IL-13 and TNF-? and up-regulate the gene expression level of IL-2,IL-10,IL-13 and TNF-? m RNA in normal lymphocytes groups and infecte d lymphocytes groups.
