Rice is one of the most important grain crops in the world,so its yield level will directly relate to food security.The yield components of rice included tiller number,spikelet number per panicle and 1000-grain weight,which were affected by many factors,such as plant height,seed setting rate,heading date,plant leaf shape and so on.The analysis of the genetic based on the important agronomic traits,such as plant height,seed setting rate and spikelet number per panicle,was greatly significant to our understanding of the composition of rice yield.In this experiment,the Indica Rice Variety "ZS97" was treated with 1%EMS?methyl ethyl sulfonate?A total of 650 M2 lines were obtained,and 36 mutant materials were screened out.Among them,1 mutants showed plant height,spikelet number and seed setting rate were significantly different from those of wild type,which was tentatively named "ZSXS".The normal seeds of M2 generation were collected from each plant,and all seeds collected per plant were planted,and M3 segregating population was obtained.Genetic analysis showed that the recessive traits were controlled by a single gene,and the gene was mapped by MutMap.The main results of this study were as follows:?1?Compared with wild type ZS97,the "ZSXS" mutant showed a shorter height,with an average decrease of 22.7cm,the spike length became shorter with average 9.0cm,the grain density became thinner,the number of branches became less at two times,the length of grain became shorter,the length and width of flag leaf became smaller,the number of grains per panicle became smaller,and the yield per plant decreased by 96.14%.The analysis of variance was significantly different from that of wild type ZS97?2?We totally got 19 candidate gene by using MutMap.The mutation loci of four candidate genes LOCOs02g12120,LOCOs03g56910,LOCOs03g59840,LOCOs04g38980 were in intron region,six candidate genes LOCOs01g15180,LOCOs10g15250,LOCOs12g16400,LOCOs03g26880,LOCOs04g52460 and LOCOs04g57100 were retrotransposons or transposons,the amino acids of the two gene,LOCOs12g31340 and LOCOs04g51840,did not change,and were samesense mutation.Through screened,the LOCOs04g39430 loci was selected as the major candidate genes.?3?LOCOs04g39430 was used as the main candidate gene locus for experimental verification.10 WTs and 40 Muttants were selected randomly,then their LOC04g39430 locis were sequenced,the data was analyzed using LightScanner.The results showed that the LOCOs04g39430 gene mutation and mutant phenotypes were isolated,All the mutant loci in short mutant plants were mutated,and the loci in the normal phenotype were homozygous or heterozygous?4?The LOCOs04g39430 gene in wild type and mutant was sequenced.Sequence alignment showed that the mutation of LOCOs04g39430 was occured at the eighth exon region by nucleotide substitution,so it leads the codon GCG mutate to ACG,and the amino acids of alanine translate to threonine.
