Fusarium head blight(FHB),caused mainly by Fusarium graminearum Schwabe,is a severe disease of wheat world wide which greatly decreases the yield and grain quality.There are two major resistance type in wheat,with type ? against the initial penetration and type ? against fungal spread within spikes.Our previous works have located Fhb5,a type ? resistance gene,on chromosome 5 A from landrace Wangshuibai.To further precisely map Fhb5,two secondary F2 populations with different genetic back grounds were developed to construct a high-resolution map of Fhb5 interval by newly developed markers based on BAC clone sequences and colinearity analysis among the wheat deletion-mapped ESTs,rice genome sequence and Aegilops tauschii(Ae.tauschii)gene map.The recombinants were genotyped by markers flanking the Fhb5 interval and the FHB resistance evaluation was carried out in two field traits in 2012.All the recombinants could be divided into two distinct classes,one is resistant to FHB and another class is susceptible.Compared the genotype and phenotype data of the identified lines with recombination occurring within the Xgwm304-Xgwwm415 interval,the Fhb5 was refined to a 0.12-0.17cM interval flanked by Xmag8217 and Xgwm415.
