Tobacco is an important economic crop in China while potato virus Y (PVY) is one of the most important pathogen in tobacco production. Recently, the PVY is outbreak of plague and increasingly serious in our country. Breeding PVY-resistance variety is the main method of PVY prevention in current production and mining the resistance gene is the key project.We demonstrated that the tobacco NtTCTP is close ralated to PVY resistance in tobacco previously.In order to study the function of NtTCTP to the PVY resistance, we carried the PVY inoculation test onthe overexpression lines and silencing lines of NtTCTP under the condition of both in greenhouse and in the field. The results showed that in NtTCTP silencing lines, both the PVY incidence rate and degree are significantly reduced, the incidence is only about 20% in greenhouse after PVY inoculation and almost no natural occurrence of PVY in the field. In contrast, the incidence rate and degree of PVY have no obvious change whether in greenhouse and in the field in NtTCTP overexpression lines. This test showed that silencing of NtTCTP in tobacco confers strong PVY resistance and NtTCTP is a new recessive gene for PVY resistance.Because of eiF4E is the only PVY resistance genes in plants, in order to comprehend the anti-PVY mechanism of NtTCTP better, the NtTCTP, eiF4E and each protein gene of PVY have been built into the yeast two-hybrid vector to verify the interactions between NtTCTP and eiF4E. each protein of PVY through the yeast two-hybrid experiment. The results showed that there is no interaction between eiF4E and NtTCTP, so the function of NtTCTP is not related with eiF4E and there may be a new anti-PVY approach, the results also showed that there is interaction between NtTCTP and CP of PVY. This study showed that NtTCTP PVY resistance is due to the interaction relationship with CP of PVY, laiding a foundation for the further study of the interactions mechanism between NtTCTP and PVY.
