Tan sheep is Chinese only production of expensive er-mao fur unique sheep variety.Because of long-term natural selection and artificial breeding,Ningxia Tan sheep with meat,hair,skin production become one of the valuable sheep variety.Tan sheep fur is known for its beauty,lightness,warmth and durability.It is different in flower form pattern in the er-mao period,and thus shows different types of spikes.However,with the age increase,the fur and other characteristics of Tan sheep will gradually weaken or even disappear.Tan sheep skin is the foundation of the fur.In order to understand internal causes the proteome can be used to study about Tan sheep skin.Based on the above background,the following was my work:Two dimensional gel electrophoresis technique was used to study about the different expression proteins in the skin of one year Tan sheep.Then skin protein map of one year Tan sheep in four growing periods was compared.19 protein spots were found,and 13 proteins of 15 protein spots were identified.The proteins related to the regulation growth of Tan sheep were 14-3-3 protein a subtype,annexin A2,keratin 25,tubulin alpha chain.The differential expression proteins were analyzed and found that these differentially expressed proteins may be related to the cyclical changes of hair follicles in different growth stages of Tan sheep skin.Isobaric tags for relative and absolute quantitation(iTRAQ)combined with multidimensional liquid chromatography-tandem mass spectrometry(LC-MS/MS)were used to study the different expression of sheep skin protein in Tan sheep,Inner Mongolia Sunitou sheep,Zhongwei goat and Xinji fine wool sheep.Proteins Discoverer 1.4 software was used for quantitative analysis,and the differential proteins were identified by database search.Finally,the GO and Pathway analysis were used to analyze the different proteins.The results showed that there were 1161 proteins in the sheep skin,and 30 different proteins were found in Inner Mongolia Sunitou sheep and Tan sheep.112 different proteins were found in Zhongwei goats and Tan sheep.107 different proteins were found in the Xinji fine wool sheep and Tan sheep.The results showed that KRTAP11.1,KRT25,KRT34 and KRT83,TCHH and KRTAP6 were related to the thickness and curvature of Tan sheep wool.In this study,the iTRAQ proteomics method and the LC-MS/MS technique were used to study the different expression of four kinds of spike-type fur-like proteins in th in the er-mao period Tan sheep of the ChuanZiHua type,RuanDaHua type,LvDouSi type,Other irregularities type.2886 proteins were identified.The number of different proteins were 135 in the RuanDaHua type and ChuanZiHua type.The difference between the LvDouSi type and ChuanZiHua type were 142,other irregularities type and ChuanZiHua type comparison to obtain the difference protein 113.The results showed that annexin and VEGF signaling pathways may be related to the hair plait formation RuanDaHua type,KRTAP3 and KRTAP6 may be related to the ChuanZiHua type hair plait structure.The above studies could provide the theoretical basis and scientific basis for the annual variation of Tan sheep skin,the breeding of high quality er-mao fur and the breeding of Tan ChuanZiHua type.
