III Maize(Zea mays L.)belongs to gramineae,zea genus,which is one of the mostimportant food crops in the world.Genetic improvement of corn is always an important issue in the field of agricultural sciences and heredity improvement method for maize is also concerned.At the present study,with the GFP corn obtained via the pollen-mediated transformation approach,we investigated the agronomic characteristics of transgenic maize and examined the genetic stability.The main results are as follows:1.Obtainment of GFP maize.Plasmids with GFP gene was introduced into maize inbred line ’ Zheng 58’ by pollen-mediated transformation method with ultrasonic aid.Kernal setting rate was 13%.Six lines of transgenic maize were obtained in total 15 corn kernals which were verified by PCR amplification.The T1 kernel positive ratio was about 40% based on harvested kernals which was 15.2.Agronomic traits analysis of of T2 plants showed the stem diameter in T2 showed significant difference compared with the control group though T2-6 was the exception.Plant height and ear height in T2-5 and T2-1 lines presented significant differences compared with the untransformed control.Overall,the main agronomic characters of T2 plants were basically identical to the control.3.Trait investigation of T2 exhibited that ear diameter was significant difference compared with the untransformed control,so was the ear length(not applied to T2-4and T2-5);ear row in T2-2,T2-5 and T2-6 showed significant difference compared to the control;Numbersof row kernel and dry ear mass in T2-1 presented differences compared to the control.Some strains reached significant difference,some strains did not reach,for example in ear length and ear diameter appeared significant difference.4.Observation of T2 plants grew in the field showed that T2-5 presented a marked variation that is,ears appeared on aerial roots and the angle between the corn cob and maize stem is about 90 o.5.The analysis of GFP gene heredity rule in T2 plants showed that T2-1、T2-5and T2-6 lines followed Mendelian genetic law.6.The results of RT-PCR analysis verified the expression of foreigner gene in varied T2 lines was not the same,which also applied to each plant from the same line.7.The western-blot analysis showed that GFP protein expression was varied among T2 plants,several lines presented GFP silence and no GFP protein expression. |
