In this paper,we used Illumina Miseq sequencing and real-time fluorescent quantitative PCR to analyse microbial communities in soil of Hippophae rhamnoides shrub,Caragana jubata scrub,Betula platyphylla forest,Quercus wutaishansea forest,Populus davidiana forest and Larix principis-rupprechtii forests of Pangquangou Reserve.Meanwhile,the activities of catalase,urease,invertase and soil physicochemical properties were measured to analyse the variation and correlation among them.To understand the ecological relationship between bacterial community and environmental factors and spatial variables in Pangquangou nature reserve,to provide theoretical basis for predicting the response of microbial community to environmental changes in Pangquangou nature reserve.We observed that:1.The urease activity was closely related to ammonium nitrogen,organic matter,total nitrogen,total carbon;The invertase activity was closely related to total nitrogen,total carbon,total sulfur and organic matter;The catalase activity was closely related to ammonium nitrogen.2.In addition to plant diversity of Caragana jubata scrub were higher in July,the other plots were higher in May.Plant diversity was closely related to total nitrogen,total carbon,urease activity,nitrate nitrogen.3.In the adjacent middle elevation,the abundance and diversity of the soil bacteria in B.platyphylla forest was higher than that of L.principis-rupprechtii forest,in contrast,the soil fungal abundance and diversity in L.principis-rupprechtii forest were higher.The soil bacterial diversity of L.principis-rupprechtii forest in high elevation was lower,but the abundance of that was higher,in contrast,the abundance of the fungi was low and diversity was high.In addition to abundance of bacteria in Caragana jubata scrub were higher in April,the other plots were higher in May.The overall trend of bacterial abundance were higher in April and in May and lower in July and in October.The bacterial abundance ofHippophae rhamnoides shrub was the lowest.The overall trend of the abundance of bacterial functional groups were the same.4.The differences of the community structure of fungi among three plots was great than that of bacteria.The bacterial community structure and diversity were closely related to total carbon,total nitrogen,total sulfur,pH and soil particle size.The abundance of bacteria was closely related to total carbon,total sulfur,organic matter,invertase activity.The invertase activity was closely related to Planctomycetes,Alphaproteobacteria,Gammaprotebacteria;The urease activity was closely related to Planctomycetes,Alphaproteobacteria;The catalase activity was closely related to bacteria.
