[Objective] In this study,the effects of high temperature stress on wheat agronomic traits,grain development,starch grain morphology,crystal characteristics and flour quality were studied in different stages of grain development at different stages through field and indoor high temperature stress.Forthemore,proteomic mechanism of starch granule binding protein were clarified in wheat grain.[Method] Xinchun 11 and Xinchun 39 spring wheat was used as experimental material,the two wheat ripening and quality types are different,and they were planted in the experiment field of the experimental institute of Shihezi University in 2015 and 2016.They were treated in different stage after anthesis,respectively.The kernel in different stage were taken as samples,and its grain filling rate and devolpment were determinated;starch granules morphology and crystal properties were observed;and the quality of the mature grains was determined.Also,the kernel devolpment,starch granules morphology and crystal properties,flour properties,rheological properties and proteomics mechanism of wheat germplasm at different temperatures after anthesis were analyzed.[The main results were as follows]:(1)1000-grain weight of Xinchun 11 and Xinchun 39 were decreased significantly on the early(T1)and medium(T2)high temperature after anthesis,and the effect of T2 treatment on 1000-grain weight was larger;There was no significant difference in plant height,bulk density,panicle length,spike number and spike number in different treatments.The grain hardness of wheat in Xinchun 11 was not significantly difference among treatments,but it was higher than that T2 treatment in Xinchun 39.(2)The swelling power at T1 treatment was lower significantly than at T2 treatment in Xinchun 11 and Xinchun39.The swelling power at T2 treatment was significantly higher than at CK in Xinchun 11.The swelling power at T1 and T2 treatment was significantly lower than at CK in Xinchun 39,but T1 treatment had a greater effect on wheat flour swelling potential.The number of descending values of Xinchun 11 was not significant in each treatment.The number of descending of Xinchun 39 wheat was significantly lower than that at CK.The effect of T1 treatment on grain protein was higher,there was a positive effect in the improvement of different index in xinchun 11.it were good for different index at T2 treatment in xinchun39.In addition to the gelatinization temperature,the measured values under the CK treatment are greater than at the T2 treatment,and the T2 treatment is greater than the T1 treatment;high temperature treatment,it have a negative effects on flour indicators in the xinchun 11,and T1 treatment More severe,the indicators of paste characteristics of the flour less than CK in T2 treatment,but it has not sensitive xinchun T1 treatment.The high temperature treatment had a negative effect on the flour pasting characteristics of xinchun 39,while the trough viscosity and gelatinization temperature were opposite in T1 and T2 treatment.(3)Under the high temperature treatment,the 100 grain weight of the two cultivars was lower than the control,and the maximum grain filling rate was about 18 DAA.T1 treatment also changed the morphologyof type A starch granules,but had little effect on B-type starch granules.Micropores were observed on starch granules surface at 10 DAA,the starch granules particle size of 15 ~ 20 DAA was significantly lower than that of the control,and the surface indentation of starch granules increased at 20 ~ 25 DAA in the A and B type starch granules in xinchun 11.The morphology and particle size of starch granules were slightly less affected at high temperature after anthesis in xinchun 39.Under different high temperature treatments,the diffraction peaks of starch granules in two variety were the same,but the peak intensity of starch granules was different,which indicated that high temperature stress did not affect the crystal type of starch granules,but might change the lamellar structure of starch granules.(4)A total of more than 2127 peptides and 892 proteins were identified in the wheat starch granule binding protein IBT quantification.The results of COG analysis showed that the function of wheat starch granule binding protein was mainly transformed,ribosome structure and biological origin,post-translational modification,protein conversion,molecular chaperone,gene function prediction,carbohydrate transport and metabolism.GO analysis shows that the molecular function is mainly involved in the catalytic activity and binding;in the cell component is mainly involved in the organelles,cell parts,cell energy;in the biological process,mainly involved in the metabolic process,the cell process.
