Northern winter turnip rape belongs to the winter type,which is an important oil and ecological crop in the northern arid area of northern China.This kind of cultivars adapt to harsh natural condition in the north,mainly have its excellent cold resistance.Cold resistance and sensitivity of varieties are closely related.In this study,strong winter(Longyou 6,Longyou 7),winter varieties(Longyou 12,Ningyou 2),semi-winter varieties(Lenox)and spring varieties(Longyou 3 and Longyou 4)were used to analyze endogenous hormones,photosynthetic characteristics,related gene expression and differential protein expression of different temperature-sensitive cultivars by physiological and biochemical methods and molecular biotechnology.The results showed that :1.The content of ABA in the leaves of winter rape increased with low temperature stress,spring rape was decreased.With the decrease of temperature,the rising range of winter rape varieties showed that strong winter varieties> winter varieties> semi-winter varieties.With the decrease of temperature GA3,IAA and ZR increased at first,and then decreased,and the spring varieties obviously decreased.With the increase of varieties temperature winter rape were decreased.The ratios of ABA to IAA,GA3 and ZR were highest at negative and low temperature,and the ratios of strong winter were most.2.The results showed that the decrease of photosynthetic characteristics Pn and Gs of winter cultivars were lower than spring varieties with the decrease of temperature,and Pn and Gs increased with the decrease of susceptibility.After chilling stress Ci significant upward trend,the performance differences between winter and spring,spring varieties of increase were greater than winter variety.3.BraVIN3,Bra VRN1-1,BraVRN1-2,BraFRI and BraVRN2 were analyzed related expression at low temperature.The winter rapeat 3 ℃ had reached the maximum,as follows: semi-winter variety>winter variety>strong winter variety.With the decrease of temperature,the related gene expression of winter varieties decreased less than semi-winter varieties.The expression of BraVIN3,Bra VRN1-1 and BraVRN1-2 were highest at 12℃,and the expression of BraFRI and BraVRN2 were smallest.The results showed that the wintering varieties was lower than semi-winter cultivars at low temperature,while the spring varieties had no strict requirements of low temperature4.Constant temperature 3℃ under the conditions of vernalization,with the increase of alternating temperature and processing time.The activities of POD,CAT,SOD,APX and the contents of free proline,soluble protein and soluble sugar were increased,and the growth rate of strong winter cultivars was higher than winter cultivars.Compared with the control,the indexes were increased in semi-winter,the physiological indexes were decreased with the increase of accumulated temperature.The effects of vernalization on physiological and biochemical indexes of different temperature-sensitive cultivars were different under low temperature stress.With the increase of treatment accumulated temperature,physiological and biochemical indexes were expressed as strong winter > winter varieties> semi-winter varieties.5.Based on identification of 16 proteins and analyzed of biological information,molecular function was mainly divided into six categories: catalytic activity,binding,protein binding,regulation of enzyme activity,nucleotide binding and transferase activity.The mainly biological processes involved were stress response,abiotic stimulation,endogenous stimulus response and carbohydrate metabolism.KEGG analysis involved in metabolic pathways had 14 categories,mainly involved in glucose metabolism,photosyntheticcarbon sequestration.Differential protein: Ribulose-phosphate 3-epimerase,Ribulose-1,5-bisphosphate carboxylase/oxygenase large subunit,Phosphoglycerate kinase,Fructose-bisphosphate aldolase,Ribulose bisphosphate carboxylase smallchain.Indicating that participated in glucose metabolism and photosynthetic carbon fixation had an important effect on vernalization.
