It is verified that some plant polysaccharide have immune regulation effects for human and animals.Malus pumila polysaccharide was polymer linked by different monosaccharides,and it is verified that malus pumila polysaccharide have effects of anti-tumor,anti fatigue,anti-inflamatory,antivirus,and oxidation resistance.However,there are not enough researches were conducted to study the immunology regulatory function of malus pumila polysaccharide.MTT method was used for detecting the cell vitality of RAW 264.7 murine macrophage that was exposed in different concentration of polysaccharide from apple.The Agilent Sure Print G3 Mouse GE V2.0(8*60K,Design ID:074809)gene expression profile chip was used to study the influences of malus pumila polysaccharide on gene expression profile of RAW264.7 murine macrophages.Western-Blot technology was used to study the influence that malus pumila polysaccharide have on expression of iNOS protein of RAW264.7 murine macrophages,at the same time,the influence of malus pumila polysaccharide on production of NO of RAW264.7 murine macrophages were studied by NO micromethod assay.The result of this research showed that: 1.The concentration of malus pumila polysaccharide of 20-80ug/m L was prone to promote the proliferation of RAW264.7 murine macrophage,but the concentration of 160ug/m L showed an inhibition effect.2.1597 differentially expressed genes were screened by gene expression profile chip finally(Fold Change>2,p<0.05),and the result of Go analysis showed that malus pumila polysaccharide may have influence on innate immune response,adaptive immune response,and even the process of inflammatory response and virus defense of RAW264.7 murine macrophages by wide-distributed target sites.3.The result of KEGG Pathway analysis showed that malus pumila polysaccharide is able to activate the immune response and promote the secretion of some cytokines and chemokines of RAW264.7 murine macrophages by TNF signaling pathway,NF-?B signaling pathway.4.RAW264.7 murine macrophage are prone to express more iNOS protein after it was treated by malus pumila polysaccharide,and it is found that malus pumila polysaccharide can enhance the production of NO in murine macrophages by promote expression of iNOS.In conclusion,the study verified that malus pumila polysaccharide is able to enhance multiplication of RAW264.7 murine macrophage,and malus pumila polysaccharide may have influences on immune response of RAW264.7 murine macrophage by activating the NF-?B signaling pathway and TNF signaling pathway.Malus pumila polysaccharide can enhance the production of NO of RAW264.7 murine macrophage by promote the expression of iNOS.
