Color hulless barleys as a special of germplasm resources,it is an important basis for cultivating new varieties of barley,but also a valuable resource for promoting the health of human beings,especially Tibetan compatriots.With the research and utilization of colored barley breeding methods and the increasing use and utilization of nutrition and processing quality,some of the colored hulless barley grains contain healthy natural antioxidant active substances.This research,seven varieties of black rice,2 parts of purple grains and 6 black rice barley varieties(strain)were used as experimental materials,according to the GB 5009.9—2016,GB 5009.88—2014,GB 5009.9—2016,Folin phenol color imetry,Aluminum nitrate and sodium nitrite complexation method and GB 5009.82—2016,the basic nutrient quality indexes and important antioxidant activity components of 15 different barley cultivars(lines)were determined,and the differences between different grain and color.Use barley varieties of Zhang Zi purple(purple tablets)and Kunlun 13(white)as the material,F2 population(266 individual plants)was obtained by hybridization,researched [urple barley barley grain color genetic law,using SSR molecular technology,look for related genetic markers,For the selection of new varieties of colored barley and the development and utilization of specialty products to provide a scientific theoretical basis and data support.The main research contents and results are as follows:1.The differences of the nutritional quality(starch,fat,dietary fiber and protein)and antioxidant activity substance(total phenol,flavonoids,vitamin E)content between eight color(seed coat)and seven white(seed coat)grain hulless barleys were determined.The results showed that:(1)There were differences in nutritional quality of hulless barley varieties(strains).The content of starch,dietary fiber protein,total phenol,flavonoids and vitamin E were 31.90% ~ 52.70%,10.59% ~15.39%,11.90% ~ 17.30%,0.23% ~ 0.35%,0.15% ~ 0.25% respectively.(2)There were significant differences in nutritional quality and antioxidant activity substances(except total phenol)of different grain color hulless barley varieties(strains).The average content of starch,dietary fiber,protein,total phenol,flavonoids and vitamin E of color varieties(strains)were 2.63%,3.66%,19.85%,11.54%,7.10% and 12.47% higher than white varieties respectively.Comprehensive comparison: color hulless barley in nutritional quality and antioxidant activity substance content is better than that of white huless barley.Visible,color hulless barley in regard to the development of functional health care products what have higher value of using.2.Da Zhangzi of purple hulless barley grain genetic characteristics were analyzed.The results showed that: the purple grain of hulless barleys(Da Zhangzi)and the white grain of hulless barleys kunlun 13 were hybrided,F1 seeds areintermediate(purple and white),F2 seeds were purple,intermediate(purple and white),white.X2 test indicated that the proportion of separate of F2 purple kernels and non-purple kernels followed the proportion of 1:2:1.So,it could be concluded that the relative character of purple kernels of Da Zhangzi was controlled by a pairs of not complete dominate genes.3.Simple sequence repeats(SSR)and bulked segregation analysis(BSA)were employed to detect molecular markers linked to purple hulless barleys gene in Da Zhangzi.The results showed that: A SSR marker GBM1092 of the 2HL chromosome was proved to link to the purple gene.The genetic distance from the color gene PlS was 18.6 cM.The results of this study lay the foundation for molecular marker breeding of barley grain color.
