Scutellaria baicalensis Georgi belongs to the genus a perennial herb,its roots are used as medicine,high medicinal value,market demand,the depletion of wild resources,and the cultivation of Scutellaria influenced by genetic and environmental factors,the quality difference is bigger,not easy to control.Through tissue culture technique can eliminate the influence caused by genetic differences,with short growth cycle,not affected by external environmental conditions and other advantages,has become one of the many ways of natural medicinal plant medicinal ingredients obtained.Among the many methods to improve the efficacy,exogenous addition of elicitors has become one of the most commonly used and effective methods.SA and MeJA,as important biological pheromones in plants,play an important role in plant signal transduction and anti stress reactions,and are also the most popular non biological elicitors of medicinal plants.In this study,the callus as materials,by adding different concentration of elicitors,biomass and content of baicalin determination of active ingredients of callus,while the PAL,the key enzyme of baicalin synthesis pathway of C4 H,4CL and CHS and the activity of antioxidant enzymes SOD,POD and CAT activity were measured,two in order from the aspects of key enzymes in the metabolic pathway and the antioxidant system preliminary study on Mechanism of elicitors on accumulation of medicinal secondary metabolites.The main results are as follows:1.Screening of different explants and different concentrations of plant hormones to determine the best explant culture of callus for the stem segment with node,the optimum culture conditions: MS+1.0mg/L 6-BA+1.5mg/L NAA,pH 5.8,culture temperature(25±1)?,dark culture.The callus growth cycle of S.baicalensis Georgi was 25 days.2.Different concentrations of SA and MeJA treatment alone,the callus growth was inhibited;the concentration of SA was 10?mol/L,50?mol/L,MeJA concentration is 10?mol/L,50?mol/L,100?mol/L dry weight was slightly higher than the control,but the difference was not significant.SA and MeJA synergistic effect,callus growth get promoted,dry weight was lower than the control,the difference was not significant.3.When the concentration of SA was 50?mol/L,the content of baicalin was the highest,which was 1.63 times of that of the control,which was significantly different from that of the control;When the concentration of MeJA was 50?mol/L,the content of baicalin was the biggest,which was 1.83 times of that of the control,which was significantly different from that of the control;In combination with SA and MeJA,the content of Baicalin in callus of A9 group(SA 100?mol/L+MeJA 100?mol/L)was the highest,which was 1.10 times of that of the control,but there was no significant difference between the control group and the control group.4.When the concentration of SA was 10?mol/L,the activity of SOD was the highest,the activity of POD was slightly lower than that of the control,and the activity of CAT was the highest at 50?mol/L;When the concentration of MeJA was 50?mol/L,the activity of POD and CAT was the largest;In combination with SA and MeJA,the activity of SOD in A4 group was the largest,and the activity of POD in A9 group was the largest,and the activity of CAT in A7 group was the largest.5.When the concentration of SA was 10?mol/L,the activity of C4 H was the largest,and the activity of PAL was slightly lower than that of the control at 50?mol/L,and the activity of 4CL reached the maximum at 50?mol/L;When the concentration of MeJA was 200?mol/L,the activity of PAL reached the maximum.When the concentration was 50?mol/L,the activity of C4 H was the largest,and the concentration of 4CL was 10?mol/L,and the activity was the largest.The concentration of CHS was 100?mol/L and the activity was the largest;The combination of SA and MeJA enhanced the activity of PAL,C4 H,4CL and CHS on the whole,in which the activity of PAL in A9 group was the largest,and the activity of CHS in A6 group was the largest,and the activity of 4CL in A7 group was the largest.6?SA alone.The content of baicalin is significantly negatively correlated with POD,positively correlated with CAT;MeJA alone.The content of baicalin was positively correlated with CAT;SA and MeJA synergistic effect,significantly positive correlation with the content of baicalin and 4CL.
