In recent years,through the constant and deep study for soybean antinutritional substance found ?-conglycinin could cause all sorts of young animals that produce allergic reaction in the diet of nutrition,and The different species of animals have different degrees of allergy.Some studies have shown that the ? subunit linear epitope of ?-conglycinin has significant differences with binding capacity of rabbit,piglets,dog,bass allergy serum.But the study for difference comparison about the ability of the ?-conglycinin alpha subunit linear epitope combined with different species of animal allergy serum is less.Bioinformatics method is used in this paper,Summarying paper has been published literature and selecting four representative amino acid sequences to compare the combined extent of different species of animal allergy serum combined with the epitope peptide by the method of ELISA and Dot hybridization is to study difference comparison of the linear epitope of soybean protein antigen ?-conglycinin and antigen epitope of its diffident specifies.Experiment ?:Research for predicting ?-conglycinin antigen epitope by BioinformaticsThere are two types of bioinformatics software namely DNAstar,the BepiPred1.0.According to the physical and chemical properties of protein such as protein antigen index,flexibility,hydrophilic,surface accessibility to predict ?-conglycinin ? subunit and ?-conglycinin ? subuni the linear epitop of ?-conglycinin?,Four representative amino acid sequences from published literature are selected.Experiment ?:comparison binding ability of ?-conglycinin with different species of animal allergy serumThis experiment is based on the principle of antigen and the reaction of antibody specificity,the synthetic peptide,as antigen,rabbit,piglets,the sensitization of calf serum as antibodies,to compare ability difference of four epitope peptide with rabbit,piglets,allergic calf serum by Elisa and dot hybridization methods,so as to compare the combining ability difference of the ?-conglycinin linear epitope and serumof different species of animal allergy research,the results show that the same animal with four different synthetic peptides combined with sensitive degree is significantly different(P < 0.05),The combining ability of rabbit serum and allergy peptides ?2 is the strongest.The combining ability of allergic piglets serum peptides with ?1 is the strongest.The combining ability of allergic calf serum and peptides ?2 peptides is strongest.The sensitive degree of combination of different species of animals and the same peptides is significantly different(P < 0.05),The combining ability of ?1 peptides combined with calf serum is the most powerful.The combining ability of ?2 peptides and piglets,allergic calf serum is relatively strong,peptides.However there is no obvious difference for the combination of ?1 and three animals allergic reactions.The combining ability of ?2and calf serum is strongest.Above all: Bioinformatics software can more rapidly and accurately predict antigen epitope region,while The combination for ability of ELISA and dot hybridization method proving four segment table a beta conglycinin peptides with three different species of animal allergy serum is significantly different.This study laid theory foundation of relative mechanism to reveal difference of different species of animals on the soybean protein antigen sensitization,also provide technical support to optimize the antigen protein detection method.
