Recently,marker assisted selection(MAS)is widely applied to research field and many great achievements were obtained.At present,some candidate genes related to lipid traits were screened out.Among them,CD36,ChREBP and SREBP1 gene arouse people,s extreme enthusiasm.Bodys of studies about CD36,ChREBP and SREBP1 gene focused on human diseases.Therefore,it is urgent for us to carry out researches of them in livestock and poultry.We adopted quantitative Real-Time PCR(qRT-PCR)to detect tissue mRNA expression levels of CD36,ChREBP and SREBP1 gene from 10-week ducks,and analysed its association with serum biochemical inidicators.In order to further explore effects of CD36,ChREBP and SREBP1 gene expression on duck primary hepatocytes and PC3 cells lipid indicators at cell level,overexpression vector of CD36,ChREBP and SREBP1 gene with label HIS-Tag were constructed to transfect to primary hepatocytes and PC3 cells.The results were as follows:1.In this study,we detected tissue expression levels of CD36,ChREBP and SREBP1 gene by qRT-PCR technique.The results showed that CD36,ChREBP and SREBP1 gene were highly expressed in abdominal fat,subcutaneous fat or liver,low expressed in gizzard and spleen.Association anlaysis of CD36,ChREBP and SREBP1 gene mRNA expression and serum biochemical indicators suggested that m RNA expression levels of CD36,ChREBP and SREBP1 gene expression were significantly positive correlated with triglyceride(P<0.01).Moreover,CD36 mRNA expression levels was significantly negative correlated with serum total cholesterol,globulin and low density lipoprotein(P<0.01).ChREBP mRNA gene expression levels was significantly negative correlated with serum low density lipoprotein(P<0.01)and was significantly positive correlated with serum total cholesterol,albumin and high density liprotein(P<0.05).SREBP1 mRNA expression levels gene was significantly negative correlated with serum high density liprotein(P<0.01),and was significantly positive correlated with serum total cholesterol and low density lipoprotein(P<0.01).CD36,ChREBP and SREBP1 gene maybe play an important role in regulating duck lipid metabolism.2.CD36-pEGFPN3,ChREBP-pEGFPN3 and SREBP1-pEGFPN3 recombined vector with HIS-tag was successfully constructed,and was used to transfect into duck primary hepatocytes to study its effect on lipid indicators.The results showed that the expression levels of CD36,ChREBP and SREBP1 gene in duck primary hepatocytes was significantly positive correlated with cell triglyceride(P<0.01).CD36 gene expression had negative association with cell total cholesterol and low density lipoprotein(P<0.01),which was opposite to SREBP1 gene.These suggested that CD36,ChREBP and SREBP1 gene had regulatory effects on the lipid synthesis of duck primary hepatocytes.3.CD36-pEGFPN3,ChREBP-pEGFPN3 and SREBP1-pEGFPN3 recombined vector with HIS-tag was successfully constructed,and was used to transfect into PC3 cells to study its effect on lipid indicators.Association analysis showed that CD36,ChREBP and SREBP1 gene expression had positive association with triglyceride(P<0.01)in PC3 cells.Moreover,CD36 gene expression had negative association with total cholesterol and low density lipoprotein(P<0.01)in PC3 cells,which was opposite to SREBP1 gene.In a word,CD36,ChREBP and SREBP1 gene maybe have regulatory effects on lipid indicators in PC3 cells,which was similar to that in hepatocytes.
