Correlation Analysis Between Polymorphism Of ESR,KISS-1,PGR And Litter Size Of Qianbei Ma Goat And Their MRNA Expressio Level Testing

Qianbei Ma Goat is an excellent local goat breed in Guizhou,which has the advantages of resistance to coarse feeding,strong resistance to disease,mildness,good meat quality,good quality of lambs and high reproduction rate.In this study,the polymorphisms of ESR,KISS-1and PGR genes in Qianbei sheep were screened by DNA mixing and direct sequencing,and the correlation analysis was carried out with the lambs.The expression of ESR and KISS-1mRNA in different tissues of pituitary,hypothalamus,uterus,fallopian tube and ovary were analyzed.The aim of this study was to investigate the polymorphism and expression of ESR gene in Qianbei sheep,Goat differences,to provide a scientific basis for improving the fertility of goats from the molecular mechanism.1.The polymorphism of ESR,KISS-1,PGR gene and its correlation with lamb shapeA SNP site,C127 T,was found at the exon 8 of the ESR gene,which caused a change in the coding of the amino acid.The number of litter size of the CC genotype was not similar to that of TT and CT(P <0.05),and there was no significant difference in Guizhou Black Goat.The intron of KISS-1 had be detected out of three SNP sites,G277 C,G436T,T486A;Three genotypes(GG,GC,CC)were found at G277 C locus in Qianbei Ma Goat and Guizhou Black Goat,association analysis showed that there was significant difference among theregenotypes of two goat population;three genotypes(GG,GT,TT)were found at G436 T locus in two goat population in litter size,association analysis showed that there was no significant difference among there genotypes of two goat population in litter size;three genotypes(TT,TA,AA)were found at T486 A locus in Qianbei Ma Goat,and two genotypes(TT,TA)were found at T486 A locus in Guizhou Black Goat,ssociation analysis showed thatthere was significant difference among theregenotypes of two goat population;three genotypes(GG,GT,TT)were found at G436 T locus in two goat population in litter size.No polymorphism was detected in the PGR gene and had no polymorphism.2.Studies on the Expression of ESR and KISS-1 Genes in Qianbei Ma GoatThe results of real-time fluorescence quantitative PCR showed that ESR gene and KISS-1 gene was expressed in both tissues of the two groups(single lamb group and multi-lamb group),and the ovaries were the highest and the ESR mRNA expression was significantly different from the other tissues(P <0.05),but no significant difference was found between the other tissues(P> 0.05).and the expression of KISS-1 gene in hypothalamus was significantly higher than that in other tissues(P <0.05)However,the expression of KISS-1gene in uterus,ovary and fallopian tube was not significantly different(P> 0.05).
