Yak occupies very important position in herders daily life on the Qinghai-tibet plateau,and yak,around the mining area polluted by cadmium(Cd2+)of Qinghai province,can enrich cadmium(Cd2+)into human beings and lead to a negative effect on human beings.Cadmium(Cd2+)pollution have a serious impact on the safety of yak meat products consumption.Therefore,it is important to study the biological metabolism mechanism of cadmium(Cd2+)in yaks.When we studyed experiment of yak kidney cells under various concentrations stress cadmium chloride,with the concentration of 0,5,10,20,30 ?mol/L,we found metallothionein gene?MT1?have obvious expressed after adding 5 ?mol/L cadmium chloride to yak kidney cells measured by fluorescence quantitative PCR technology,and heavy metal cadmium can promote MT1 mRNA expression.And the MT1 gene mRNA expression quantity increased while the strength of the heavy metal ion cadmium increased.It is determined that 5 ?mol/L cadmium chloride is the minimum amount of addition in cadmium(Cd2+)toxicology experiment for the subsequent trials.When analysed the genome of yak liver tissue,experimental group feeding with 3mg/ml cadmium chloride and blank group,we found 16029 differentially expressed genes measured by lllumina Hiseq2000 sequencing system,including 5596 genes up-regulated and 10249 genes down-regulated.And we found 384 significant different gene,including 129 genes significantly up-regulated and 255 genes significantly down-regulated.Compared to the blank group,we know heavy metal cadmium impacted the expression of gene a lot.When analysed the genome of yak liver tissue,we found 3173 proteins were identified to have a biological function,including 104 expressed different proteins.When analysed the proteome and transcriptome correlation of yak liver tissue,we filtrated 14 proteins through 384 significant different genes and 104 significant different proteins.The 14 proteins are isocitrate dehydrogenase,BOLA class I histocompatibility antigen,ADP-ribosylation factor,haptoglobin,dimethylaniline monooxygenase,estrogen sulfotransferase,carboxylesterase3,UDP-glucuronosyl transferase,carbonyl reductase,annexin A2,glutathione S-transferase,aflatoxin B1 aldehyde reductase,farnesyl pyrophosphate synthase,fatty acid-binding protein.
