At present,the wheat is an important food crop that is cultivated area of about 2.44 × 105 hm2,and 84% are mainly distribution in the Great Wall to the south of Henan,Shandong,Hebei,Jiangsu,Sichuan,Anhui,Shanxi province an-d so on,north of 16% distribution they Heilongjiang,Inner Mongolia,Gansu,Xin-jiang,Ningxia,Qinghai province in china.But for a long time,the wheat have been seriously impacted on the yield,quality,taste due to the harm of plant virus disease.Barley yellow dwarf virus(BYDV)disease is a more serious diseas-es,in Qinghai,studies of wheat yellow drawf disease reported less.So this stu-dy is the evaluation of wheat germplasm resources resistance to Barley yello-w dwarf virus of Qinghai province;the major epidemic strain around the Qing-hai province BYDV-GAV gene sequencing,and multiple and the sequence,clu-stering analysis and evolutionary tree built with MEGA software(MEGA,A-merican)adjacent method(Neighbor-joining),it serves as the pathogenicity o-f BYDV,host interactions and resistance to lay the foundation For later study.This study obtained conclusions as follows:1?414 wheat germplasm resources were artificially inoculated with viruliferous wheat aphid by hill plot technique in the field.The results showed that Gan A100,Chuan 766,Shan 1059,Lanmai-2,Huimutou,Lvjiankou,Mosha,Jia-mabaimangmai and Kequn filling stage of disease index were15.33,23.87,23.64,24.53,14.76,22.65,23.67,20.59,21.46,less than 25,9 copies of resistant germplasm resources and less production losses;The symptoms of 13 th cultivars,GanA-101,GanA99,Zang515,Zang519,Muhanmai,Qunkedabaimai,Lataibanmai,Xiaohongmai-2 and Langxianzheda2,Nicaibimang,Muhanm-ai,Gamaizi,Tiexiaomai,Jointing stage of the disease index is greater than the filling stage of the disease index,and the 13 varieties during the jointing stage to filling stage improvement or disappear.It was showed that 13 germplasm resistance to dise-ase.Remaining 392 filling stage of germplasm resources of disease index were higher than 25 to performant ahout disea-se or Gao Gan,and production loss is bigger.The more resistance to the yield loss is smaller,and vice versa.2?Population genetic variation of BYDV-GAV species was analyzed,the results showed that:(1)?The present of wheats which come from Xining,Haixi,Guide and Xuhua and be susceptive to yellow dwarf disease is 95.83%.(2)?There had 23 bases were mutated in 331 bases,which T?G and C?G transversion reached 62.97%,in which are dominant mutation type.It was illustrated BYDVcp bases have preferences,in the process of mutation to transform mechanism.172 protein have 7 amino acids encoding a protein nonsynonymous mutation happened,the non synonymous substitution rate was 4.07%.(3)? The four isolates are from the same one and its have similarity sequence up to 95.3%~99.9% and they are close to BYDV-GAV which in china.(4)?The Ka/Ks ratio is less than 1,indicating that evolution is stable,and BYDV-GAV is a negative choice in qinghai and is in a state of purification.
