Trichinosis caused by Trichinella spiralis,is an important food born zoonotic disease.Trichinellosis serious harm,not only caused huge economic losses to the livestock industry,but also is a major potential threat to meat food safety,is serious to human health.Now,immune control for Trichinosis is not good,because the pathogenesis and escape host immune surveillance mechanism of Trichinella spiralis are not clear.Research on parasites show that Th17 and Treg cells play an important role in clearing the parasites,inducinge inflammation,reducing pathological damage and reach parasitic balanced state between parasites and host.So,it is necessary for us to clarify which way have Trichinella spiralis utilize to maintain immune balance state between itself and host.Therefore,this study aims to discuss the influence of Th17/Treg cells on the activation and proliferation of the host that infected by Trichinella spiralis,and the effects of Th17/Treg cells on intestinal inflammation,which can analysis the interaction between Th17 and Treg cells preliminary.First of all,50 SPF BALB/c female mice aged 4-6 weeks old have divided into 2 groups randomly,each mice in experimental group were given 400 of Trichinella spiralis muscle larvaes through mouth,and the control group given equal amount of PBS.After 7 days post infectioin,we make lymphocyte suspension by mesenteric lymph nodes.Activation and proliferation of Th17 and Treg cells was detected by Flow cytometry,and Real time fluorescent quantitative PCR was used to detect the dynamic change level of IL-17,IL-6,TGF-?,IL-10 and transcription factor Foxp3,ROR? t.Finally take the duodenum to make HE staining,observe the changes in inflammation.This study showd that the number of Treg cells in Trichinella spiralis group were higher than the control group during the infected,and showd immunosuppressive action from fourteenth days.Th17 cells differentiate rapidly in the 7th day of Trichinella spiralis infection and inhibit the differentiation and development of Treg cells,the mature and proliferation of Th17 cell has been strengthen on the fourteenth day.The function of Th17 cells was decline and inflammation levels returned to normal on the thirty-fifth day under the immunosuppressive action of Treg cells.In summary,Th17 inhibit the differentiation of Treg on the seventh day,and then Treg geta dominant position to inhibit the immunologic function of Th17 owing to play immune suppression gradually which reduced the inflammatory response of intestinal,this processes also advantageous to establish the parasitic balance between Trichinella spiralis and host.
