GH?GHR?JAK2 Gene Cloned,tissue Expression And Alternative Spliceosome Analysis In Weining Goat

6-month years old,one years old,two years old of male and female Weining sheep as the research objects,the experiment used molecule methods to cloned CDS region of relation genes about JAK/STAT signal transduction(GH,GHR and JAK2),meanwhile to analysis biological information.In addition,the experiment used real-time q PCR method to analysis m RNA experssion rule of GH,GHR and JAK2 in heart,liver,spleen,lungs,kidneys and longest back muscle in different growth phases of different genders,to obtain m RNA experssion rule of GH,GHR and JAK2 in Weining sheep.The result is as show:(1)The experimnent successfully obtained the whole CDS region of GH and GHR genes from Weining sheep,respectively654 bp and 1905 bp,encoded 217 amino acids and634 amino acids.Meanwhile long of the cloned GHR gene is diferent with long of the reference sequcence in NCBI(1839bp),as a result of the there exon(66bp)spliceosome.(2)Though blast software,in GH gene we found there mutation,respectively G135 C and G646 C,G135C and G646 C of which is missense mutation,resulted in Gln?His and Pro?Ala;in GHR gene exsited T60 C and A837 T,one former be sense mutaion,the latter be missense mutation,resulted in Phe?Leu.Further bioinformatics analysis shown that single nucleotide mutation may result in secondary structure and tertiary structure change of growth hormone and growth hormone receptor protein.(3)The result shown expression level of GH,GHR and JAK2 genes in Weining sheep all have different degree of expression.The expression level of GH gene in six tissues of Weining sheep is higheast in spllen,followed by the lung,kidney,liver and longest back muscle and heart tissues;GH gene in different gender and different growth stages of the same organization has some significant differences in expression,and with the increase age,relative expression of GH gene shown the tendency of fallen slightly.The expression level of GHR gene in six tissues of Weining sheep is higheast in longest back muscle,followed by the lung,kidney,liver,spllen and heart tissues.in the heart,lungs,liver,and the longest muscle of male Weining sheep,the experssion level of GHR gene is higher that in female goat,but lower that in spllen.GHR gene expression in different growth stages of the same organizations have certain difference,and as increasing age,the expression did not show obvious regularity.expresssion level of JAK2 genes is higheast in the liver,followed by the longest back muscle,the lowest in the heart;.in the heart,liver,spleen and kindey of male Weining sheep,the experssion level of JAK2 gene is higher that in female goat,but lower that in longest back muscle,but in difference growth phases the expression level of JAK2 gene has no obvious significence,which result from gene function and individual difference.
