Salix linearistipularis is small trees that belongs to salix of Salicaceae.Due to it is a strict dioecious plant,it is important experimental material for study of male and female flower development of woody plants.Salix linearistipularis not only has strong resistance to high temperature,cold,salt and alkali,but also play an important role in wind and sand fixation.at the same time,it is the good material of landscaping.Male and female plants often have different economic value,so gender identification,especially early sex identification that is great significance for production planning and economic benefits of Salix linearistipularis.In this study,we used the female flower buds,male flower buds,female leaf buds and male leaf buds in the early stage of spring as the experimental materials,and used the Illumina 2000 for sequencing.Female flower buds,male flower buds,female leaf buds and male leafbuds were sequenced,to explore the Unigenes expression characteristics of male flower buds and female flower buds and male and female strains,lay the foundation for the study of flowering mechanism of Salix linearistipularis and early male and female identification.The results are as follows:1.Transcriptome data evaluation: 41095 Unigene and 44835 transcripts were obtained after splicing assembly.The BLAST software was used to compare the constructed transcriptome with NR and other databases and the preliminary bioinformatics analysis was performed on the transcriptome.There were 28457 Unigenes annotations on GO term,in which the cellular components,molecular functions,and biological processes were 14,15,and 23,respectively.There were 15411 unigenes showing significant similarity to the sequences in the COG database and clustered for 25 categories,there are 20456 Unigenes notes to 357 KEGG pathway.2.The co-expression of female flower buds and male flower buds:We take the leaf buds as control and finally we obtained 357 unigenes of co-expression of male flower buds and female flower buds,and a total of 37 co-expression of male flower buds and female flower buds were obtained by cluego analysis,among them twenty-five unigenes were associated with flower development,such as MADS-box,AP1,SEP2,ARF,YABBY,WUS,NEYA1,PETAL LOSS,Cytochrome P450,basic / neutral invertase,plant protein kinase and hormone-related unigenes,and to lay the foundation for future study.3.The specific expression unigenes of female flower buds and male flower buds:?The specific expression unigenes of female flower buds?We take the leaf buds and male flower buds as control and finally we obtained 292 unigenes of specific expression of female flower buds,and a total of 10 specific expression unigenes of female flower buds were obtained by cluego analysis,and 8 high flodchange and expression unigenes from the specific expression unigenes of female flower buds.?The specific expression unigenes of male flower buds?We take the leaf buds and female flower buds as control and finally we obtained 153 unigenes of specific expression of male flower buds,and a total of 8 specific expression unigenes of male flower buds were obtained by cluego analysis,and 3 high flodchange and expression unigenes from the specific expression unigenes of male flower buds.4.The specific expression unigenes of male and female individualsIn order to obtain the specific expression gene of male leaf buds and female leaf buds,we obtained 33 unigenes of female leaf buds compared to male leaf buds,and 8 unigenes of male leaf buds compared to female leaf buds;In order to obtain a marker gene that is more likely to identify sex,we obtained 1362 unigenes of female flower buds compared to male flower buds,and 1215 unigenes of male flower buds compared to female flower buds,finally,we get 9 unigenes which are expressed specifically in female plants?7 unigenes expressed specifically in male plants,which lays the foundation for obtaining the marker unigenes that can do the sex identification of Salix linearistipularis at early stage.In summary,we obtained co-expression unigenes of female flower buds and male flower buds?specific expression unigenes of female flower buds and specific expression unigenes of male flower buds from the analysis of Salix linearistipularis of female flower buds?male flower buds and leaf buds,these unigenes provided important information for further understanding of the flowering mechanism of plants,male and female differentiation,and early sex identification.
