| The hypovirus/C.parasitica system is an excellent model system for the study of interaction between virus and host.This system has already been applied on virus-related bioresearch for target genes and host factors.Autophagy is highly conservative in all eukaryotes.Its function and dysfunction play very important roles in microbial infection,aging and neurodegeneration.The atg8 mutant strains not only displayed significantly reduced virulence,but also decreased virus accumulation after infected by hypovirus CHV1-EP713.The recombinant plasmid pCPXG418-atg8-RFP was constructed and transformded into protoplasts of pCPXHY2-p40-GFP/EP155,pCPXHY2-p48-GFP/EP155 and pCPXHY2-hel-GFP/EP155.Transformants of pCPXG418-atg8-RFP/pCPXHY2-p40-GFP,pCPXG418-atg8-RFP/pCPXHY2-p48-GFP and pCPXG418-atg8-RFP/pCPXHY2-hel-GFP were obtained successfully.Compared with the wild type strain EP155,the transformants didn’t show significant changes in phenotypes when grew on PDA plate.Autophagy protein Atg8 was found to be co-located with viral proteins P48,P40 and Hel in autophagic vacuoles using fluorescence microscope.This study provides some basis on the interaction between autophagy protein Atg8 and viral proteins P48,P40 and Hel. |
