In order to determine the mechanism of eugenol anesthesia carps,to investigate the effect of NMDA receptor agonists and antagonists on the effect of eugenol anesthetized carps,the changes of NMDA receptor antagonists on the eigenvalue curve of eugenol anesthesia carps.The effects of eugenol on the expression of excitatory and inhibitory amino acid neurotransmitters mechanism weather the effects of eugenol anesthesia on the expression of NMDA receptors in the central nervous system of the carps.To improve the safety of fish anesthesia,reveal the mystery of the brain function of the fish and the development of new,high efficiency and low toxicity fish provide a theoretical basis for general anesthetic.1?48 cyprinoid carps,each weight between 700~800g as the experimental animals were randomly divided into one blank control group and five anesthesia groups.The anesthesia group was intraperitoneally injected with 100 umol/L NMDA receptor antagonist MK-801,each 0.3 ML,and the control group was injected with the same volume of saline.After the injection into the water for 10 s.In different concentrations of eugenol solution the carps were anesthesiaed,the study of NMDA receptor antagonist on eugenol anesthesia carp effect.The results showed that NMDA receptor antagonist could accelerate the time of anesthesia and prolong the awakening time,and the number of chewing in the anesthetized carp was significantly decreased(p<0.05),and the number of times of movement in the air per minute was significant decreased(p < 0.05),and the time of exposure to exposure to air in the air was prolonged.Therefore,the NMDA receptor antagonist had the effect of enhancing the eagle anesthesia of the carps.2?40 cyprinoid carps as the experimental animals were randomly divided into 5anesthesia groups and eight carps in each group.The anesthesia group was injectedintraperitoneally with 100 umol/L NMDA receptor agonist Spermine,each 0.3 mL,and the control group was injected with the same volume of saline.After the injection into the water for 10 s.In different concentrations of eugenol solution the carps were anesthesiaed,the study of NMDA receptor agonists on eugenol anesthesia carp effect.The results showed that agonists of NMDA receptors could reduced eugenol anesthesia effect in carp.Carps could enter anesthesia for a long time,recovery time faster.And the number of cheek movement of the carps decreased significantly(p<0.05),the number of bouncing increased significantly in the air per minute(p<0.05),carp start the time to shorten the spring from the water exposed to the air.3?64 cyprinoid carps as the experimental animals were randomly divided into eight anesthesia groups and eight carps in each group.The study the changes of the dose-response curve in the normal time after the anesthesia group and the eugenol anesthesia antagonist group and the carp anesthesia.Results: The dose curve of NMDA receptor antagonist + MK-801 was not parallel to that of eugenol,the Dose-effect curve of NMDA receptor antagonist + MK-801 and eugenol curve starting point can the same,and the slope and maximal have was changed,indicating that eugenol and +MK-801 acted on different receptors,NMDA receptor was not site of action of eugenol anesthetized fish.4?21 cyprinoid carps as the experimental animals were randomly divided into one blank control group,three eugenol anesthesia control groups,three NMDA receptor antagonist groups and three carps in each group.The method was characterized by high performance liquid chromatography(HPLC)changes in excitatory Amino Acids and inhibitory Amino Acids.The results showed that the contents of glutamate and glycine in the cerebellum and spinal cord were decreased in the NMDA receptor antagonist groups compared with the control group in the optimal anesthetic concentration range,while the content of aspartic acid and ?-aminobutyric acid of the content were increased.It's indicated that NMDA receptor antagonists enhanced the effect of eugenol anesthesia carp,maybe through the cerebellum and spinal cord excitatory amino acids and inhibitory amino acid content.5?21 cyprinoid carps as the experimental animals were randomly divided into one blank control group,three eugenol anesthesia control groups,three NMDA receptor antagonist groups and three carps in each group.Application of high performance liquid chromatography method studied the cerebellum changes in excitatory Amino Acids and inhibitory Amino Acids.The results showed that the content of aspartate,glycine and ?-aminobutyric acid was increased in the range of the optimal anesthesia concentration,and the content of aspartate,glycine and ?-aminobutyric acid was increased in the NMDA receptor agonist group compared with the control group.While the content of glutamate and gamma-aminobutyric acid in the spinal cord decreased,the content of aspartic acid and glycine increased.
