Three homologous genes of Arabidopsis type-B response regulators(ARRs)were isolated from soybean cultivar Tianlong 1 and named as GmARR10-A(Glycine max ARR10-A),GmARR10-B and GmARR10-C.To investigate the biological functions of GmARR10 s,plant expression vectors 35S::GmARR10-A/-B/-C were constructed and transformed into Arabidopsis and soybean cultivar Tianlong 1.The results are as follows:1.The coding sequences of GmARR10-A,GmARR10-B and GmARR10-C are 2034 bp,2097 bp and 2040 bp,repectively,all of which are completely coincidence with that of the reference genome Glycine max Wm82.a2.v1.2.Five Arabidopsis transgenic plants overexpressing GmARR10-C were obtained by floral dipping.When treated with 200 nM 6-BA,transgenic plants exhibited much shorter roots compared with that of wild type plants.But the roots of transgenic plants without 6-BA treatment were obviously longer than that of wild type plants.3.After drought treatment for 3 weeks,the survival rate of transgenic plants overexpressing GmARR10-C was 23.24%,compared with 29.42% of wild type plants.When treated with 50 mM NaCl for 8 days,the survival rate of transgenic plants was 27.98%,compared with 95.79% of wild type plants.Furthermore,the expression levels of GmARR10-A,GmARR10-B and GmARR10-C in soybean were reduced after drought and salt treatmert.4.Three transgenic soybean plants were obtained by cotyledonary node transformation,which provided the materials for functional research of GmARR10 s.5.To investigate the biological function of GmAP1.2,the plant expression vector p35S::GmAP1.2:GFP was constructed and transformed into soybean cultivar Tianlong 1.Three transgenic plants were obtained and used to analyze the phenotypes during growth and development.Our research showed that overexpression of GmAP1.2 in soybean induced early flowering about 31 days,the decreases of plant height and leaf area,and increases of Chlorophyll a,Chlorophyll b and Chlorophyll contents by 1.43,1.52 and 1.57 times respectively.However,floral organs of transgenic soybean were normal.The results suggested that GmAP1.2 could play roles in flowering regulation,plant architecture,leaf development and the chlorophyll accumulation in soybean,but not in floral organ development.
