Avian Salmonella infection has a serious impact on chicken production and it is also an important zoonosis,which involves food safety.In present study,serological investigation and analysis of Salmonella infection in Yantai,Weihai and Weifang were conducted,and the isolation and identification of Salmonella were made in order to provide scientific basis for the control of Salmonella infection in regional population.1.The epidemiological investigation of salmonella in breeding birdsThe separation rate of Salmonella was 3.8%from 91 batch of flocks of the environment,chicks,hatching eggs and other 107422 samples continuously from 2012-2015 in 55 chicken farms.Among all batchs,42 of different age The positive rate of Salmonella averaged plate up to 21.3%at 19 weeks and 10.3%at 58 weeks,and the average plate aggregation of 23 batch to the highest rate at 19 weeks,reaching 7.52%,the same age as broiler chickens,other age are below 6.8%,the positive rate of broiler chickens was significantly higher than the egg breed;classify chicken flocks according to the annual of introduction,the positive rate of plate agglutination of Salmonella showed a decreasing trend every year.The plate agglutination rate after 8-fold dilution of serum was more obvious.The positive rate of plate agglutination of Serum from 30-50 weeks of age in the ancestors of 2015 was 0%,but there was an upward trend after 55 weeks,the positive ratet show a rend of decline year by year after 36 weeks.However,However,in the case of serum 8 times dilution,the positive rate of plate agglutinationt was significantly lower than that in 2013-2014.Peak positive rate from 2%to 10%of the interval,down to 2%or less,but still higher than the end period of ancestral broiler breeder egg production.2.The undiluted positive serum of salmonella and 8 times dilution of the flat plate were still positive serum detection rate and specific comparison.Salmonella continues to be isolated from the hatching eggs,the chicken serum 8 times diluted salmonella plate agglutination still positive serum,with BIOCHEK B + D group Salmonella antibody detection kit for detection,positive self-agreement reaches 97.9%,and 98.8%of the pathogens were isolated from the eggs,the two methods have a high correlation.However,there was a significant difference between the positive group of the undiluted serum flat-panel and the 2-fold dilution of the serum.The ELISA method was used to detect the positive serum of 317 flat-pack positive serum in 5 fields that were not isolated to salmonella pathogen.There was a significant difference in the positive rate between the positive field of a meat breeding and a negative field of a pathogen,but the negative field also had persistent plate agglutination-positive production;eight times dilution plate agglutination positive only in positive pathogen separation field detection,eight times diluted plate agglutination positive more conducive to eliminate the interference of false positive evaluation does wild infected chickens,easy to take corresponding control measures.3.Identification of salmonella isolatesA total of 67 strains of Salmonella isolated from different fields and different sources were selected from 2012 to 2016,and 59 strains of Salmonella enteritidis,Salmonella typhimurium 1 strain,suspected Serratia strains 1,Arizona 6 strains were isolated by PCR,biochemical reaction and single factor serum agglutination.Isolates were mainly Salmonella enteritidis.The tracking of the positive variety of chicken farms found that the death rate of the chickens in the whole feeding cycle was significantly higher than that of the negative chickens.The isolation rate of Salmonella typhimurium was 0.05%-0.33%in chicks.Breeding of chickens concentrated infection,resulting in the next generation of chicks premature rush,the complaint rate increased.4.Prevention and Control of Salmonella in Breeding FarmThrough the development of HACCP(Hazard Key Point Control Program)program,strengthen biosecurity measures,feed control,field pest control,disinfection of water and litter,acidizing agent use,incubation hygiene control,explosion,the use of Salmonella enteritidis vaccine immunization,regular detection and elimination of positive chicken and other comprehensive measures,the production capacity of laying chickens was significantly improved,the complaint rate has decreased significantly.
