Potatoes(Solanumtuberosum),the world's fourth most important food crops which after wheat,rice and cron,is solanceae perennial herb and can be eaten.China is the world's largest potato production country,followed by Russia,India and Ukraine.In 2015,China launched the strategy of potato staple food to promote the potatoes into bread,noodles,rice and other staples.So potatoes will become another staple of rice,wheat and maize.But in the process of potato plant,it faces many threats of disease such as scab disease of potato,bacterial wilt,dry rot,early blight and late blight.Among them,the potato scab disease is most serious.Once potato sufferied the scab disease,the appearance of potato will be seriously affected,and this diease may even cause potato yields lower as well as impede the potato's commercial marketing.The disease is a soil-borne disease caused by Streptomyces,and pathogenic Streptomyces produce metabolites thaxtomins.Typically,thaxtomins can lead to dark brown,corkysuberin on the surface of potato,forming bumps scab,depression or netted scab.S.scabies,S.turgidiscabies and S.acidiscabies are three of the main potato scab pathogen.Microbes play a very important role in the process of biological coversion of organic matter and biological control of agricultural production.With the development of molecular biologe,based on 16 S r DNA molecular biology technique,such as DGGE,T-RFLP,played a very important role,above all,HTS makes it more and more easier in the stduy of microbial diversity.In this experiment,it analyzed the microbial diversity of scab lesion of potatoes with common scab disease by using the method of Illumina High-throughput sequencing and the method of traditional culture.The results are as follows:(1)During the mothed of HIS,it finds 21 bacterial phylum,47 bacterial class,139 family and 210 genus.It also finds 4 fungus phylum and 76 fungus genus.Derived from microbial composition OTU that,Streptomycetes and Fusarium account for the highest proportion,followed by Flavobacterium.(2)By the mothed of unculture,it find 109 isolates,among of them,it have 42 strains of Streptomycetes,49 bacterium and 18 fungi.The main strains on the face of ill potatoes are Streptomycetes,Pseudomonas,Flavobacterium,Agrobacterium and Fusarium.The possibility of pathogens is Streptomyces acidiscabies.(3)Correlation analysis of species shows that,Acinetobacter has the similar changce with the possibility of pathogens.So it can be guess that Acinetobacter may have the relation with the pathogenesis of potato common scab.Agrobacterium,Flavobacterium and Sphingobacteria has the antagonistic effect with Streptomyces acidiscabies.So it provides the basis for the treatment of potato common scab.(4)The interaction between strains s hows there are antagonism effect between Pseudomonas mediterranea and the possibility of pathogens.
