Malus sieversii was lived in China's Central Asia Xinjiang Tianshan,which is the ancestors of apples.and his values has been attention by many people.But in recent years,there are a lot of problem around it.in a word,The exploitation and utilization of Malus sieversii resources are imminent.FLS2 was an important component of the plant immune system,which was related to the resistance of plants such as invasion of foreign pathogens,activation of downstream resistance genes.It could also cause the initiation of plant's MAPK and the activation of ROS.In a word,it played an important role in the stress response of plants to pathogens.Although the research of FLS2 functional mechanism in Arabidopsis thaliana was relatively clear,but many researches on the FLS2 in apples were still in the initial stage because of the complexity of its own genes.So,it was hard to find more domestic researches on the FLS2 in apples.In this study,we cloned the FLS2 gene in the ‘Malus sieversii' apple and transfered its gene into the Arabidopsis thaliana,then made some researches on the heterologous expression in Arabidopsis thaliana and some of phenotype in Arabidopsis thaliana.At the same time,we constructed the prokaryotic expression vectors of FLS2 and BAK1 kinase regions and purified the corresponding proteins.We got some results,which laid the foundation for the study of the function of FLS2 in apples and the deeper works in the future.The main results of this study were as follows:1.The fragment size and corresponding sequence of FLS2 in apples were determined.The structure of the protein was analyzed by bioinformatics,and the function of FLS2 in apple was determined.2.MdFLS2 overexpression in Arabidopsis thaliana reduced the seed germination rate of Arabidopsis thaliana,and the plant growth of Arabidopsis thaliana was slowed down.3.Overexpression of MdFLS2 partially restored susceptibility to Arabidopsis thaliana mutant FLS2 to bacterial flagellin.4.The prokaryotic expression vectors of MdFLS2 and MdBAK1 kinase regions were constructed and the proteins of MdFLS2 and MdBAK1 kinase were successfully induced and purified,which laid the foundation for the experiment of other proteins.
