Diarrhea in piglets is a common disease in the production process.There are many factors causing diarrhea in piglets,among which bacterial diarrhea is one of the important pathogens affecting the healthy growth of piglets.In this study,the feces of piglets with diarrhea were collected from large scale pig farms in Lanzhou City,and were sent to the laboratory for isolation and identification of pathogens and molecμLar biological identification of pathogens identified by bacteriology.The aim of this study was to confirm the main pathogenic bacteria caused by large-scale pig farms in Lanzhou City,and to select reasonable antibiotics through drug sensitivity test,so as to provide a theoretical basis for clinical prevention and treatment of piglet diarrhea.Select four large-scale pig farms,120 fecal samples of piglets with diarrhea were collected.From May 2015 to August 2016,smear microscopy was performed first,and then Eosin Methylene Blue agar medium and Salmonella were selected.CμLture medium,blood agar plate,SS,MacConkey,nutrient agar plate,modified CCDA agar,brain heart infusion blood agar medium and other special cμLture media for pathogen isolation and purification;biochemical identification of isolated strains,and then select 20 susceptibility papers such as ampicillin,cefazolin,and cefuroxime were used for drug susceptibility testing;the isolated strains were subjected to 16 S rRNA gene amplification and sequencing,and blast alignment was performed in GenBank to analyze the homology of the bacteri.The resμLts showed that: in this study,112 Escherichia coli strains were isolated from the collected materials in this study.108 strains of Clostridium viridis;53zhu;96strains of jejunal bendriosis;13 dead Bacillus strains;The resistance rates of Escherichia coli and Keleibojun to cefazolam,cefotaxime,cefotaxhene,ampicronin and other drugs are relatively high,and the drugs such as imamipenem,cefpiperazone,and Pailaxilin are sensitive.The resistance rates of Clostridium Weisporis relatively high to ampicillin and Clostridium are relatively high.Sensitivity to most other antibiotics;The isolated strains diagnosed by bacteriology extracted DNA for PCRamplification,and the obtained target gene fragment sequencing results were compared in GenBank and then compared with known E.coli,Clostridium Weiscens,Crabella,Aspergillus jejunum and necrosis.Sequence has a high homology.The isolation and identification of pathogens for collecting diarrheal diseases of piglets requires the selection of a reasonable cμLture medium.Staining microscopic examination combined with biochemical tests can comprehensively determine the types of pathogenic bacteria;PCR detection can increase the diagnostic specificity,sensitivity and accuracy of pathogens,and gene sequencing can be accurately known.The type of bacteria can shorten the time for diagnosis;the drug sensitivity test can detect the degree of drug resistance of bacteria and improve the basis for clinical drug use.
