Ammopiptanthus mongolicus is a kind of shrub which is extremly drought tolerant,it is the ancient subtropical evergreen broad-leaved forest in Tertiary age that experienced the shrink and drought in Mediterranean sea.What's more,it is the only evergreen broad-leaved fruticose flora in our northwest area.Amomopiptanthus mongolicus not only can fix sand,afforest the mountains and be admired by people,but also it has great pharmacological values.Although there has been some researches that showed the seed of the Ammopiptanthus mongolicus was antiviral and immune,there isn't any authoritative report to prove its antioxidatioxidation and immunocompetent around the world.The further explore on its functions is so important.This thesis analyses from these aspects:?1?The acute toxicity test using total flavonoids after purification?86.78%?.?2?The study on Antioxidant activitity of the total flavonoids by using Hydroxyl radicals?·OH?produced in invitro Fenton system and the superoxide radicals?O2-?produced by pyrogallol autoxidation to determine the scavenging effect of total flavonoids.At the same time,using D-galactose to replicate mouse aging model,the effects of total flavonoids on serum malondialdehyde?MDA?content and superoxide dismutase?SOD?activity in aging rats were examined.?3?The satitistical analysis on the dynamic changes of intraepithelial lymphocytes and goblet cells by giving the mice total flavonoids of Ammopiptanthus mongolicus at different concentrations for 15 days and then taking duodenum,jejunum and ileum to make routine paraffin embedded sections.The experiment results:?1?In the acute toxicity of total flavonoids?86.78%?of Ammopiptanthus mongolicus seeds,no animal died at the test when the dose is above?5000mg/kg.It met the standards of non-toxic compounds such as"Food Toxicology","Pharmacological Experimental Methodology"and"Animal Toxicology".?2?The total flavonoids of Ammopiptanthus mongolicus seeds has good scavenging ability to·OH and O2-.At the concentration of 1250?g/mL,the scavenging effects on·OH and O2-were best,and the clearance rates respectively reached96.45%and 50.39%.?3?Total flavonoids of Ammopiptanthus mongolicus seeds could significantly increase the activity of superoxide dismutase?SOD?in mouse serum.Compared with the aging model group,the high,medium,and low doses of the three groups could significantly increase the activity of SOD?P<0.01?,in which the medium-dose group?150mg/Kg·d?has the best effect on SOD activity,up to 102.42±2.11(U·mL-1).?4?The total flavonoids of Ammopiptanthus mongolicus seeds could significantly reduce the serum malondialdehyde?MDA?content in mice.Compared with the aging model group,the three groups of high,middle and low doses can significantly reduce the content of malondialdehyde?MDA?in serum of mice?P<0.01?,and the middle group?150mg/Kg·d?had the most significant reduction,reaching 15.16±0.27(nmol·mL-1).?5?Total flavonoids of Ammopiptanthus mongolicus seeds could significantly promote the proliferation of mouse intestinal mucosal intraepithelial lymphocytes?IEL?and mucous goblet cells.Compared with the blank group,the highest dose group?250mg/Kg·d?could significantly increase the number of IEL and goblet cells in duodenum,jejunum and ileum?P<0.05,P<0.01?.In conclusion,the total flavonoids of Ammopiptanthus mongolicus seeds is non-toxic compounds and has good antioxidant activity.In addition,it can induce the proliferation of murine intestinal mucosal immunocompetent cells.
