Yak(Bos grunniens)is a unique species of livestock on the Tibetan Plateau in China.Ingestion of rough pastures has greater damage to the gastric mucosa,so the pre-gastric mucosal epithelium has different degrees of keratinization.Abomasum is the only stomach with genuine digestive significance.As a food storage and processing plant,it can digest and absorb the nutrients it takes,which is of great significance to the survival of calves.At present,domestic and foreign scholars have studied the histological structure of yak abomasums.However,there are few studies on the histological structure of yak abomasums stomach-secreting cells.There is no report on the expression and quantitative study of GSDMA and HSP70 in yak abomasums.The purpose of this study was to study the histological characteristics of the endocrine cells in the abomasal gland of Yak,and the distribution and differential changes of expression of heat shock protein GSDMA and HSP70 in the the rumen,the reticulum,the omasum and the abomasum of yak.The gastric tissue of 8 adult healthy yaks was selected and the type of cells in the abomasal gland was studied by histological staining.The distribution and expression of GSDMA /HSP70 in different stomach were detected by immunohistochemistry and Western blotting,respectively.GordonSweet staining showed that a large number of endocrine cells were found in the abomasal gland.The results of AB-PAS coloration revealed that the endocrine cells had different properties in the abomasal gland.Immunohistochemistry showed that the positive expression of GSDMA was only found in the epithelial cell layer of yak,a mong them,the stratum corneum cells showed strong positive expression,Some transitional cells between granular layer and stratum corneum are not positively expressed,The basal and spinous cells are also positive expressed;The scope of the abomasum positive reaction was very wide,not only expressed in the epithelial cells of the mucous surface,but also expressed in the endocrine cells,chief celland and parietal cells in the abomasal gland.Immunohistochemistry and Western blotting results showed that the highest expression of HSP70 was found in the abomasal of Yak,and the expression in rumen,reticulum and omasum was descended in turn.Among them,the expression of GSDMA protein in the abomasum was significantly different,from that in the rumen,reticulum and valve(P<0.01),and there was a significant difference between the rumen and the reticulum and valve-stomach(P<0.05).The difference was not significant(P>0.05).Immunohistochemistry showed that the positive expression of HSP70 was only found in the epithelial cell layer of yak,but strongly positive in the stratum corneum cells,while some cells in basal layer,granular layer and spinous layer were moderately positive expressed,whereas some other cells were negative.The scope of the abomasum positive reaction was very wide,not only expressed in the epithelial cells of the mucous surface,but also expressed in the endocrine cells and parietal cells in the abomasal gland.Meanwhile,the positive reaction was mainly concentrated in the cytoplasm and nucleus of the cells.Worthy of our attention,immunohistochemistry and Western blotting results showed that the highest expression of HSP70 was found in the abomasal gland of Yak,and the expression in rumen,reticulum and omasum was descended in turn(P<0.01).This experiment provided basic data for further understanding the histological characteristics of yak abomasum and the distribution characteristics and expression changes of GSDMA/HSP70 in yak stomach.
