| ThTIP and ThDUF106 genes were involved in the stress tolerance of Tamarix hispida,and the expression of ThTIP and ThDUF106 was up-regulated in Tamarix hispida under salt-alkali stress.In order to investigate the function of Th TIP and ThDUF106 genes in Tamarix hispida,the temporal and spatial expression of these two genes were detected by qRT-PCR in the leaves and roots of three-month-old Tamarix hispida under abiotic stress such as salinity,drought and heavy metals.The genes encoding these two genes were also analyzed,and the two genes were constructed and transformed into tobacco and 84K poplar(Populus alba×P.grandulosa‘84K Yang’)by the plant expression vector.The results showed that:(1)The expression patterns of ThTIP and ThDUF106 genes in leaf and root under abiotic stress were analyzed by QRT-PCR.The two genes were expressed in leaf and root,however,the expression patterns were different under different stress.The expression levels of these two genes were increased under 150μl ClCd2 and 0.4 M NaCl,respectively;(2)The full length of ThTIP and ThDUF106 genes were 759bp and 741bp,respectively,which were cloned by PCR and cloned into yeast expression vector(pYES2).The transgenic yeasts were treated with salt stress,drought stress and heavy metals,and the yeast growth status was observed.Transgenic yeast showed enhanced tolerance to the above abiotic stresses;(3)Two genes were transformed into pROKII,andintroducedintotobaccobyAgrobacteriumtumefaciens-mediated transformation.The transgenic tobacco plants and the wild-type seedlings were treated with saline,alkali,drought and heavy metals.The physiological and biochemical indexes The results showed that transgenic tobacco and non-transgenic tobacco under the stress conditions didn’t exhibit significant difference in stress tolerance,suggesting overexpression of two genes in tobacco did not improve tobacco resistance;(4)When two genes were transformated into 84K Yang poplar,the results showed that the expression of ThTIP and ThDUF106 genes in 84K poplar did not improve the stress resistance of transgenic plants.The above results indicated that ThTIP and ThDUF106 genes responded to stress in Tamarix spp and yeast,but their overexpression did not improve the tolerance of transgenic plants to tobacco and 84K Yang. |
