Ocimene can improve the resistance of plants,but the ocimene signal is through which mechanisms to regulate the expression of related defense genes in plants to improve plant resistance,so far there is no in-depth study.In this study,Pattern of biological Arabidopsis,monocotyledonous plant corn,dicotyledon rapeseed were used as experimental materials.The only feeding cruciferous plants Plutella xylostella,omnivorous Prodenia litura used as test insects.Through the methods of insect resistance,single factor experiment and RNA-seq,the molecular mechanism of ocimene promoting plant insect resistance was studied,and the following test results were obtained.1 The treatments were used ocimenene to treat corn,rape,Arabidopsis thaliana,respectively,and then inoculated with Plutella xylostella and Prodenia litura.It was found that the insect-resistant ability of corn,rape and Arabidopsis thaliana treated with basilene was improved,and the rate of Antifeedant index(AFI)and Inhibition rate of weight gain was significantly increased compared with the untreated group.Among them,when treated with 5 ?M of Ocimene and treated with 2-year-old Prodenia litura in corn,the AFI of the larvae was 33.61 ± 3.86% and the Inhibition rate of weight gain was 62.95 ± 5.98%.When treated with 5 ?M of Ocimene and treated with 2-year-old Plutella xylostella in rape,the AFI of larvae was 52.56 ± 5.25% and the Inhibition rate of weight gain was 32.05 ± 3.49%.;when treated with 5 ?M of Ocimene and treated with 2-year-old Prodenia litura in Arabidopsis thaliana,The AFI of larvae was 44.03 ± 9.65%,and the Inhibition rate of weight gain was 59.46 ± 4.71%.2 The single factor experiment was used to identify the optimum conditions for the improvement of insect resistance of Arabidopsis thaliana.The minimum treatment time was 5?M,the shortest treatment time was 3h,the shortest reaction Time 3h.Under these conditions,the AFI(%)of the abusers rate was 43.12 ± 3.98%.3 The Arabidopsis thaliana seedlings were treated with the optimal optimum conditions for Ocimene treatment,and the untransformed Arabidopsis seedlings were used as control.Sequencing of transcription.The results of the sequencing were analyzed.The results are showed on the behind.(1)23,931,053 clean reads were obtained,and further 23837 genes were obtained.GO and KEGG pathways were annotated for all genes,of which 8388 were annotated to the GO database and 10122 genes were annotated into the KEGG database.(2)Gene expression analyses showed that 88 Arabidopsis thaliana was significantly differentially expressed in the untreated group(FDR?0.001 and the factor of more than 2 times),and 75 genes were up-regulated and 13 genes were down-regulated.In the differential gene function annotation obtained,differentially expressed genes focus on defense and resistance,protein and amino acid synthesis and other functions.There are 27 genes that are a directly related to plant defense and resistance,including functions such as organic matter,inorganic matter,hormones,stress,and other biological responses.Among them,there are 26 genes directly associated with protein and amino acid synthesis,including P450,VSP protein and so on.In addition,there are 42 genes associated with additonal protein synthesis,and the expression products of these genes may contain specific proteins related to resistance,which may be involved in the basala-promoting plant resistance to insect resistance.(3)Selected AT3G28740 and other genes to do qPCR validation,the results and RNA-seq basically the same.These results show that Ocimene promotes plant resistance to insects and is likely to be directly related to the pathways of defense and resistance to plants,and also serve as related to the synthesis of specific proteins related to resistance.This study provides a lot of reference data for further understanding the molecular mechanism of Ocimene promoting plant insect resistance.
