WRKY contains a large family of transcription factors that play an important role in stress response and plant development,while the role of most of WRKY in strawberry is not well known.Here,FvWRKY71 was isolated and cloned from woodland strawberry ‘Ruegen'.Transgenic plants and transcriptome sequencing analysis provide some information for the in-depth study on FvWRKY71 function in strawberry.1.The coding sequence of FvWRKY71 gene is 1107 bp.Subcellular localization analysis showed that FvWRKY71 was localized in the nucleus.Transactivation analysis assay displayed that FvWRKY71 had transcriptional activation activity in yeast.2.Expression profiling revealed that FvWRKY71 was expressed in all tested strawberry organs,especially in red fruits.The transcript level in the red fruit stage was about 6 times than that in green fruit stage,while the expression level was low in roots,petioles,leaves and flowers.3.We overexpressed FvWRKY71 in Arabidopsis and strawberry,three over-expressed Arabidopsis lines,four over-expressed transgenic strawberry lines,and six RNAi-transgenic strawberry lines were obtained.Phenotyping indicated that transgenic plants showed early flowering and increased branching compared with wild type.4.A lot of differential expressed genes related to flowering and branching were identified by RNA-seq analysis of wild-type and overexpressing FvWRKY71 Arabidopsis plants.The hormone-related genes LBO1,AHP6,GA20ox2,GA3ox1,YUC1,CPK6,and flowering related genes such as LFY,FPF1,FUL,SPL3,and FLC were screened to provide research ideas for shortening the flowering time of Rosaceae.
