Objective:To explore effects of ephedrine on EPI, E2concentration in blood, dishistologicalstructure, activity of superoxide dismutase (SOD) and maleic dialdehyde (MDA)content in ovary of mice and research the effects of Chinese medicine on ovary inmice of ephedrine injury; the change of ovarian histological structure were observedof filial mice, to study the effects of ephedrine to filial mice development.Methods:Kunming mouse108accorded to ratio of1:2of male and female were bought,Caged together, checked the vaginal plug or did a sperm smear observed sperm underthe microscope in the next day, denoted as1d of pregnancy. Pregnancy mice weredivided into control group, ephedrine group(2g/L?4g/L?6g/L) and Chinese medicinegroup?The ephedrine group intraperitoneal injected escalation doses (2.0?4.0?6.0g/L)of ephedrine after pregnancy for5days. The control group were injected with saline,Chinese medicine group were injected with0.2ml the ephedrine of6.0g/L and werefilled with0.3ml of Chinese medicine after injecting ephedrine1hour, until the15dafter childbirth, free food. Using ELISA method to detect concentration changes ofEPI and E2in plasma, the changes in the enzymatic activity of SOD and MDAcontentwere respectively analyzed by colorimetry, the change of ovarian histologicalstructure were observed by microtechnique, counted the change of numbers ofsecondary follicle and atretic follicle by Image pro plus6.0, measured the expressionof Bax protein and ER-? in the ovary were measured by immunohistochemistrymethod.Results:1. Ephedrine affected hormone levels in plasma of mice. After the injection ofephedrine, ephedrine group EPI content of mice compared with the control groupwere increased with injection of ephedrine doses rises, significant difference or verysignificant(P<0.05or P<0.01). The E2concentration in Plasma increased with theinjection of ephedrine reduced, significant difference or very significant(P<0.05or P< 0.01).2. Ephedrine affected the antioxidant enzymes activity on ovary of mice. Afterinjection of ephedrine, activity of SOD in ovarian in each experimental group werelower compared with the control group, and the dose-related. Ovarian MDA contentin each experimental group were higher compared with the control group,4g/L and6g/L ephedrine group MDA content of ovarian compared with the control group thedifferences were significant or highly significant (P <0.05or P <0.01).3. Ephedrine affected ovarian weight and ovarian histological structure of mice.After the injection of ephedrine, the ovarian wet weight of experimental groupdecreased compared with the control group, significant difference (P <0.05). Therewere pyknotic ovarian tissues, including oocyte ateophy, degradation granulose cells,and karyopyknotic granulose lutein cells in the experimental groups. The numbers ofprimary follicle, secondary follicle and atretic follicle in each experimental groupscompared with the control group increased, significant difference or very significant(P<0.05or P<0.01).4. Ephedrine could affect the weight of filial mice and ovarian? theorganizational structure of ovarian?After the injection of The mice which inpregnancy?the weight of filial mice and ovarian in every ephedrine group werelower compared with the controlled ones?which had significant or very significantdifference?sP<0.05or P<0.01??the ratio of primordial follicle in ovarian of5d filialmice was significant lower?and the structure of secondary follicles in10d and15dfilial mice was less clear?the granular cells were scattered?5. Chinese medicine affected the antioxidant enzymes activity on ovary in miceof ephedrine injury. Chinese medicine has a significant impact on the content of EPIin plasma of mice of ephedrine injury, EPI content in Chinese medicine group weresignificantly or very significantly lower than the ephedrine group (P <0.05or P <0.01),the content of E2increased compared with the ephedrine group, significant difference(P<0.05). Ovarian SOD activity were higher compared with the ephedrine group,significant difference (P<0.05). Ovarian MDA content were decreased, significantdifference or very significant (P<0.05or P<0.01).6. Chinese medicine affected ovary weight and the organizational structure in mice of ephedrine injury. Chinese medicine after administration, ovarian weight ofmice increased compared with the ephedrine group, significant difference (P <0.05);the structure of ovarian was clear, there were a structured of cumulus, granulosa cellsclosely arranged, disintegration were reduced; the numbers of secondary follicle andatretic follicle were decreased compared with the ephedrine group, the diameter ofsecondary follicle were very significantly increased (P <0.01).7. Chinese medicine affected expression of Bax protein and ER-? in ovary ofmice ephedrine injured. After giving Chinese medicine, expression of Bax protein inovarian of mice decreased compared with the ephedrine group (P <0.05, P <0.01), theexpression of ER-? in ovaries of ephedrine group increased, the expression of ER-? inovary of Chinese medicine group decreased, each time period differences were highlysignificant (P <0.01).Conclusion:1. After the injection of ephedrine, There were pyknotic ovarian tissues, fuzzystructure, the numbers of primary follicle, secondary follicle and atretic follicle wereincreased, the diameter of secondary follicle were decreased, the numbers of corpusluteum were decreased, corpus luteum were damaged, the secondary follicle oocyteshrinkage, granule cell degeneration and degradation. Ephedrine had great effect onthe ovarian histological structure and development, seriously damaged the ovarianhistological structure and function of mice.2. Ephedrine as adrenoceptor agonists, enter the body of mice, competitivelybinded adrenoceptor, agitated receptor at long time and made it Internalized anddegradation, the content of EPI in plasma of mice increased. Ephedrine could reduceactivity of ovarian SOD, destruct antioxidant enzyme systems and make content ofMDA in ovary increased, caused by nucleic acid and protein of ovarian granulosacells, luteal cells crosslinking. Affected ovarian follicular development in mice,leading to apoptosis or karyopyknosis and content of E2in plasma reduced.3. Ephedrine could affect the weight of filial mice and ovarian?Ephedrinecould enter filial mice through maternal placenta or milk?inhibiting the food consumption of Filial mice and the absorption of the ileum increasing catabolism?making the weight of filial mice and ovarian reduced? 4. Ephedrine could affect the organizational structure and development ofovarian?Ephedrine could increased the reserves of primordial follicle of filialmice?promoting the primordial follicle development into primary follicles?obscuring the structures?diffusing the arrangement of granular cells?5. The active ingredient in Chinese medicine contained these effects: sedation,reducing the excitability of amphetamine the central nervous system neurotransmitter,regulating concentration of Ga2+, oxidation resistance, inhibiting the expression ofBax protein and reducing the ratio of Bax/Bcl-2and other. The mice were protectedby Chinese medicine that had lower content of EPI, lower autonomic activities, higheractivity of SOD and lower lipid peroxidation levels. Ovarian atrophy slightly ease inmice and ovarian histological structure be more clear, secondary follicle and atreticfollicle were increased, luteum karyopyknosis and granulosa cells disintegration werereduced, the expression of Bax protein were also reduced. Show that Chinesemedicine could protect ovarian ephedrine injuried of mice.
