The Screen Of The Parasite-derived Molecular To Evaluate The Severity Of Liver Pathology In Host With Schistosomiasis Japonica

Objective To select the molecular to evaluate the severity of host liver pathogenesis after Schistosoma japonicum infection and investigate its mechanism.Method The levels and the titers of GST-Sj HSP40-Ig G,Sj HSP70-Ig G,and Sj HSP60-Ig G specific antibody in infected mice serum and rabbits serum were determined by ELISA,and the correlation with the extent of the hepatic pathology in infected mice,rabbits and patients were analyzed by Spearman.We immunized the C57/BL6 L mice with the fusion protein respectively.Then,the titer of Ig G antibody in mouse serum was determined by ELISA,and the proportions of Th2,Th17,and Tfh cells in total CD4+ T cells were analyzed by FACS respectively.Results Compared to controls,the levels of GST-SjHSP40-IgG,SjHSP60-IgG,and Sj HSP70-Ig G antibodies were increased in infected mice and rabbits.Moreover,the level of GST-Sj HSP40-Ig G,Sj HSP60-Ig G,and Sj HSP70-Ig G antibodies in infected mice with injection of CCl4 were higher than those in the infected mice without injection of CCl4.The titers of GST-Sj HSP40-Ig G and Sj HSP70-Ig G antibodies had no correlation with the area of the live single egg granuloma in infected mice.However,the titer of Sj HSP60-Ig G antibody was positively correlated with the area of the single egg granuloma in infected mice live.The titers of Ig G anti-GST-Sj HSP40 and Ig G anti-Sj HSP70 antibodies had no correlation with the grades of collagenⅠ、collagen and Ⅲα-SMA expression.However,compared to the controls,the expression levels of α-SMA and collagen Ⅲ,not collagen expression Ⅰlevel,were positively associated with the titer of Sj HSP60 Ig G antibody in infected rabbits.After immunization of mice with GST-Sj HSP40 、 Sj HSP60 、 Sj HSP70 corresponding fusion protein,we found that either GST-Sj HSP40,Sj HSP60,or Sj HSP70 fusion proteins markly induced the production of Ig G specific antibody compared to PBS control groups.The results showed that either Sj HSP40,Sj HSP60,or Sj HSP70 had no remarkable influence on the frequencies of Th2 and Th17 cells in total CD4+ T cells.However,only Sj HSP60 induced the increase of the Tfh cells.Furthermore,our investigation in schistosomiasis patients in endemic area showed that the titer of Ig G anti-Sj HSP60 antibody was positively related to the extent of hepatic pathology.The proportion of peripheral blood Tfh in schistosomiasis patients was significantly higher than normal person.Conclusions Our results suggest that SjHSP60 protein can be used as a diagnostic molecule to assess the level of hepatic pathology in schistosomiasis patients.
