Schisandra was recognized as a new type of "medicine food homology" functional health food, variety of active ingredients was contained, but also has strong pharmacological activities. This study was rsearched to separate Schisandra chinensis fructus and seeds, the effective components were extracted fully, and the biological activity of effective components was determination. The experimental results wew as follows:(1) The lignans of Schisandra chinensis seed were extracted by ultrasonic extraction method, and the optimum process was getted: ethanol volume fraction is73%, ultrasonic extraction time was 32 min and solid-liquid ratio was 299 mL·g-1, th extraction rate was 2.47%.(2) The content of lignans of Schisandra chinensis seed was optimized by composite enzyme, the optimum process parameters were: neutral protease 5.0%,laccase 1.0%, cellulase 0.5% and pectinase 0.5%, pH6.0, enzymolysis time 30 min,temperature 30?. Under this condition, the extraction yield of Schisandra chinensis seed was 3.91%, was 1.58 times than that without enzyme technology(2.47%).(3) The total phenols of Schisandra chinensis fructus wew extracted by ultrasonic extraction, the best extraction technology conditions as follows: 44%ethanol concentration, ultrasonic extraction time for 15 min, liquid-solid ratio was 215ml·g-1; In this condition, the extraction yield of total phenol was 1.19%.(4) The total phenols were extracted by composite enzyme method, the optimum process parameters were optimized as follows: mannase 1.0%, cellulase 3.0%, neutral protease 3.0%, laccase 5.0%, enzymed time was 20 min, pH6.0, the temperature was30 ?, under this condition, the extraction yield was 2.13%, was 1.8 times than that without enzyme technology(1.19%).(5) The total phenols of Schisandra chinensis fructus were confirmed of having strong scavenging effect on the DPPH free radicals, the concentration of 1.0 mg·mL-1,the scavenging rate was compared with vitamin E and BHT, asically reached 100%,and the clearance rate of samples rise speedly, samples has a strong antioxidant capacity.(6) High performance liquid chromatography(HPLC) results showed that the total phenols substances were included the gallic acid, catechin, chlorogenic acid,epicatechin, rutin and ferulic acid. The rutin was the highest content, and that gallic acid, rutin were confirmed to have the main composition of antioxidant activity.(7) 10 different extracts were determined the inhibition rate by ?- glycosidase enzyme, seeds of petroleum ether layer IC50 value was 3.412 mg·mL-1, has good effect on blood glucose, and lower than acarbose IC50 value(3.718 mg·mL-1), the seeds of petroleum ether layer was determined of very good prospects in inhibiting alpha glycosidase enzymes.(8) 10 different extractions were measured NO2-clearance, the seeds of methanol extraction on NO2- clearance were the best. The inhibition rate was 83.4% of 20mg·mL-1 oncentration. The different layers were extracted on NO2- clearance of IC50 value was: the seeds of methanol layer > the seeds of ethyl acetate > the seeds of water layer > the seeds of methylene chloride layer > the seeds of petroleum etherlayer, the IC50 value of seeds of methanol layer was 2.159 mg·mL-1. The clearance rate of fructus of methanol layer increased with the concentration, but slowly close to flat, while the fructus of water ether layer was still rising, others not. The IC50 value of the fructus of methanol layer was 2.025 mg·mL-1, below the seeds of methanol layer, and also lower than Vc(IC50 value of 3.267 mg·mL-1), it was showed that fructus of methanol layer was the strongest removing.In conclusion, the extraction yield of fructus and seeds were increased by enzymatic extraction, Different parts and different solvents extraction, have different pharmacological activities.
