ObejectiveTo study the expression and biological significance of HIF-1?, VEGF-C and VEGFR-3 protein in prostate cancer and normal tissues, and analysis of the relationship among expression of HIF-1?, VEGF-C and VEGFR-3 in prostate cancer tissues. To study the possible mechanism of prostate cancer. Method89 cases were obtained from Department of pathology, the Affiliated Hospital of Yan'an University with complete clinical data during 2006- 2013, among in 64 prostate cancer and 25 normal prostate tissue specimens. After HE staining by the hospital pathologist diagnosis, all patients did not undergo chemotherapy or radiotherapy. The age of patients with prostate cancer ranged from 47 to 86 years old, average 68. In 64 cases of prostate cancer classification, the score of prostate cancer Gleason pathology according to the 2004 version of WHO urinary and male reproductive tumors, 18 cases was 2 ~ 4, 27 cases was 5 ~ 7and 19 specimens was 8 ~ 10; According to the 2002 AJCC TNM standard for clinical staging: 41 specimens was T1 a ~ T2 c and 23 specimens was T3 a ~ T4; According to the lymph node metastasis: 40 cases had no lymph node metastasis and 24 cases with lymph node metastasis. The expression of HIF-1??VEGF-C and VEGFR-3 protein was assayed by means of immunohistochemical technique. By SPSS software package, ? 2 in spection, statistical analysis the expression HIF-1?, VEGF-C and VEGFR-3 in prostate cancer tissues; The correlation among HIF-1?, VEGF-C and VEGFR-3 by using Spearman rank correlation analysis, there were significant differences for P<0.05. Result1. The expression of HIF-1??VEGF-C and VEGFR-3 positive material was tan-yellow granular. The expression of HIF-1? positive was in the nucleus and the cytoplasm, and VEGF-C and VEGFR-3 protein was mainly in cytoplasm.2. The expression of HIF-1? protein of prostate cancer in the Gleason score, with the development of cancer cell Gleason score level from low to high, the positive expression of HIF-1? protein increased gradually, there was significant difference between the scores of Gleason(?2=2.855,P<0.05); The positive expression of HIF-1? protein in TNM clinical stage T1 a ~ T2 c group was 48.8%(20/41) and in T3 a ~ T4 group was 73.9%(17/23). There was significant difference between of them(?2=3.816,P<0.05); The positive expression of HIF-1? protein with the lymph node metastasis was 91.7%(22/24)and without the lymph node metastasis was 42.5%(17/40). There was significant difference between of them, too(?2=15.233,P<0.01).3. The expression of VEGF-C protein of prostate cancer in the Gleason score, with the development of cancer cell Gleason score level from low to high, the positive expression of HIF-1? protein increased gradually, there was significant difference between the scores of Gleason(?2=2.692,P<0.05); The positive expression of VEGF-C protein with the lymph node metastasis was 100%(24/24)and without the lymph node metastasis was 52.5%(21/40). There was significant difference between of them(?2=16.213,P<0.01). The positive expression of VEGF-C protein in TNM clinical stage T1 a ~ T2 c group was 65.9%(27/41) and in T3 a ~ T4 group was 78.3%(18/23). There was no significant difference between of them, too(?2=1.087,P>0.05).4. The positive expression of VEGFR-C protein with the lymph node metastasis was 100%(24/24)and without the lymph node metastasis was 55.0%(22/40). There was significant difference between of them(?2=15.026,P<0.01). The expression of VEGFR-C protein of prostate cancer in the Gleason score, with the development of cancer cell Gleason score level from low to high, the positive expression of HIF-1? protein increased gradually, there was no significant difference between the scores of Gleason(?2=1.562,P>0.05); The positive expression of VEGFR-C protein in TNM clinical stage T1 a ~ T2 c group was 70.7%(29/41) and in T3 a ~ T4 group was 73.93%(17/23). There was no significant difference between of them, too(?2=1.087,P>0.05).5. To analysis by Spearman rank of HIF-1?, VEGF-C and VEGFR-3 in all cases of prostate cancer tissues, the expression of HIF-1? was positive correlation related with VEGF-C and VEGFR-3 positively, so dose VEGF-C and VEGFR-3 in prostate cancer tissue. The rs was 0.685 between the expression HIF-1? and VEGF-C(P<0.05), and the rs was 0.701 between the expression HIF-1? and VEGFR-C(P<0.05) and the rs was 0.831between the expression VEGF-C and VEGFR-C(P<0.05). Conclusion1. The expression of HIF-1?, VEGF-C and VEGFR-3 in prostate cancer overexpress.2. The expression of HIF-1? in prostate cancer have significantly associated with pathological grade, clinical stage, lymph node metastasis, that prompt the process of prostate cancer's occur, development and invasion is closely related to the anoxic factor HIF-1?.3. The expression of VEGF-C in prostate cancer have significantly associated with pathological grade and lymph node metastasis, but no significantly associated with clinical stage, that prompt VEGF-C induced lymphatic hyperplasia in the process of lymphatic metastasis in prostate cancer. VEGF-C can be used as a test for prostate cancer malignant degree index.4. The expression of VEGFR-3 in prostate cancer have significantly associated with lymph node metastasis, and no significantly associated with the score of Gleason and clinical stage, that prompt VEGFR-3 have formation of the promotion of the role of lymphatic vessel.5. There is a positive correlation among HIF-1?, VEGF-C and VEGFR-3, that suggest the three interaction and jointly promote the occurrence of prostate cancer.
