Ultrasound contrast agents are used to enhance the backscattering of soundwaves of the blood vessels and to improve the resolution of the diagnosis.Currently, microbubble contrast agents such as Sono Vue are widely used in echocardiography. Microbubbles have the size about 1-4?m and cannot be used for selective tumor imaging due to that the size is larger than the leakage of 380-780 nm in the tumor blood vessels due to the rapid vascular formution. Nanoscale contrast agents, on the other hand, have advantages by enhanced permeation rention in the tumor vasular structure with its smaller size.The goad of this study is to establish a targeted ultrasound contrast agents, which could provide a basis for early diagnosis of cancer. The main contents are:1) Optimization of nanobubble formula with 0.42% L-polylysine NB,with the sizebetween 200nm-300 nm, enhanced ultrasound images.2) Established Bel7402 subcutaneous tumor model to provide a good model for thesubsequent experiments.3) Fluorescence imaging indicated that concentrated in the tumor at 0.5h, then reachthe highest fluorescence intensity at 2h. After 24 h, the fluorescence intensitygradually decreased via the kidneys and the bladder excretion.4) Ultrasound imaging indicated that significantly enhanced tumor peripheral contour.
