Objective:A large number of studies have indicated that the polymorphism of Apo E gene is associated with various diseases such as coronary heart disease and other diseases.Statins can reduce blood lipidand provide resistance against inflammation Using high fat diet building atherosclerosis rats model, and the model rats were given atorvastatin for the treatment, the determination of rat Apo E gene expression changes as well as the changes in LDL-C, HDL-C, TC, TG, C-reactive protein and atherosclerosis index.To investigate the relationship between the changes of Apo E gene expression and the statins. So as to guide the clinical intervention of high-risk genotypes of coronary heart disease, reduce the incidence of cardiovascular events,reduce the economic burden of the family. Methods: 30 SD rats were divided into three groups:(A) control group(B) experimental group(C) intervention group: each of them is 10. A group were given basic feed, B, C group were fed with high fat diet to manufacture atherosclerosis rats. After modeling for 8 weeks, each group kill 2 rats and observe the atherosclerosis. from the 9th week C group rats were given atorvastatin 5mg/kg/d, A, B group were given the same amount of saline, gavage for 4 weeks. Then kill all the rats and Determination of TC, TG, LDL-C, HDL-C, C reactive protein in rats and the expression of Apo E in rat liver by real-time PCR, calculate the Atherosclerosis Index. Results:(1)Compared with the control group, experimental group and intervention group, serum TC, TG, LDL-C increased significantly(P<0.01).(2)The experimental group compared with the intervention group and intervention group, serum TC, TG, LDL-C decreased(P<0.05),and the HDL-C increased(P<0.01)(3)Compared with the control group, the experimental group rats Apo E m RNA expression decreased(P<0.05);(4)the experimental group and the intervention group compared to intervention group rats PCR expression was increased obviously(P<0.01).(5) CRP were higher in the experimental group and the intervention group(P<0.01) in the intervention group CRP were lower than the experimental group(p <0.05);(6)the experimental group compared with the control, AI in experimental group was significantly higher(P<0.01),(7) the intervention group compared with the experimental group, AI in the intervention group was lower(p<0.01)(8)the intervention group compared with the control group shows no significant difference(p>0.05) Conclusion: Atorvastatin can reduce the level of serum TC, TG, LDL-C, Apo E m RNA, increase the HDL-C and expression of Apo E m RNA in the liver of atherosclerotic rats. Reduce the conversion of cholesterol, and untilmately achieve the goal of reducing the risk of atherosclerosis. C-reactive protein is association with atherosclerosis, it can be ragard as an independent predictor of coronary heart disease...
