Objective:To observe the therapeutic effects of moxibustion on experimental RA bone destruction;To explore the mechanism of moxibustion in the treatment of osteoclasts in bone destruction in RA.Methods:Thirty japanese big ear rabbits were randomly divided into three groups:RA model group、control group、moxibustion treatment group、each group of 10 animals. Model group and moxibustion group with Freund’s adjuvant arthritis model. within 7 days after the building began to respectively with moxibustion applied to Japanese big ear rabbit "shen shu","zu san li" hole,1 times a day,each time treat for half an hour,6 days for a period of treatment,the total of 5 treatment cycles. With rabbits right knee joint bone tissue (retained cartilage part) retained,observing the animal sections after been HE stained;Using Immunohistochemical staining SP method count rabbit osteoclasts and real-time fluorescence quantitative PCR(RT-PCR) expression of OPG/RANKL protein gene determination.Results:1.Pathological morphology:RA model group compared with the control group:Articular cartilage damage is very serious, cartilage surface roughness, the transitional layer fracture can be seen, cartilage samples completely stripping (P<0.01), articular chondrocytes sequence out of order, four layers of structure resolution is not clear (P<0.01), tidal line break (P<0.01),trabecular derangement, trabecular area narrow, empty bone lacuna increased, part of the small beam fracture, the osteoclasts were increased; The moxibustion group compared with the RA model group, cartilage the structure breaking bad light, that owe smooth cartilage chondrocytes (P<0.01),four layer structure is clear, Only a few inflammatory cells in articular cartilage, articular cartilage damage slightly (P<0.01), tidal line the faintly visible (P<0.01),the trabecular mild disorders, a small amount of empty bone lacuna, trabecular no fracture.2.The number of osteoclasts:RA model group compared with the control group RA rabbits articular bone osteoclast count were significantly elevated (P<0.01); moxibustion treatment group compared with the RA model group RA bone osteoclast count reduced significantly (P<0.01).3.(1)Gene expression of RANKL:the model group compared with the control group RA rabbits articular bone RANKL expression were significantly increased (P<0.01);moxibustion group compared with the RA model group, the expression of RANKL was significantly lower (P<0.05).(2)The expression of OPG gene:RA model group compared with the blank control group, the expression of OPG gene was significantly reduced (P<0.01); moxibustion group compared with the RA model group, the expression of OPG gene elevated significantly (P<0.01).Conclusion:1.Moxibustion can reduce the experimental RA rabbit bone destruction and delay the progress;2.Moxibustion could inhibit RA in rabbit articular overexpression of RANKL, enhance the expression of OPG, limit the number of osteoclast over expression.
