There are four medicine centipedes in traditional Chinese medicine,which Scolopendra multilans are most extensive in clinical application.Centipede as a treament for the disease has a very long history.Long ago,centipe is known to have been used for treating snakebite,sore,spasm,apoplexy and other diseases.Now people also found it has anti-tumor,anti-bacterial,anti-aging and so on.The bodies of Scolopendra mutilans as the raw material were extracted with methanol and alcohol,then isolated with the silica gel column chromatography,ODS column chromatography.At last,four compounds were obtained.On the grounds of chemical and spectroscopic methods,their structures were identified as Uracil(1),Taurine(2),4-Hydroxy-benzoic acid(3),Quinoline-3,8-diol(4).We determine the content of Quinoline-3,5-diol and Quinoline-3,8-diol in Scolopendra mutilans,using HPLC,with acetonitrile-water as mobile phase,gradient eluting,Ultravidet absorption detection wavelength of 240 nm.Experimental results show that Scolopendra multilans(Anguo City in Hebei province),Quinoline-3,5-diol content is 0.92 mg/g,Quinoline-3,8-diol content is 1.70 mg/g.We use 7-Chloroquinoline as basic sheletons,methods of organic synthesis and NMR technology as a tool,combined with biological activity tests,carried out research work to optimize its strueture.By 7-Chloroquinoline at 3 position and 4 position structural modification and transformation,exploration and research synthesis route,the final 12 Quinoline derivatives was synthesized.By biological activity tests,we found that a series of synthetic compounds have better anti-AChE activity.
