FGF-2 Promotes The Level Of Synaptophysin On The Endogenous Neural Stem Cells In The Mouse Model Of ALS

Abstract/Summary:
Background:Amyotrophic lateral sclerosis(ALS)is a kind of adult onset progressing and fatal neurodegenerative diseases,mainly involving spinal anterior horn cells,brainstem motor nerve nuclear and cerebral cortex vertebral somatic cell,which specific etiology and treatonent,so far,is not clear.Endogenous neural stem cells exist in the central nerve system of the adult animals and human,which are in static state in normal,these cells will be activated to migrate and differentiate into some types of neurons in the majority of neural system diseases,this method and mechanism is still not clear.Fibroblast growth factor 2(FGF-2)one of main neurotrophic factors,has been proved to have a role of promoting endogenous neural stem cells.Objective:To detect the improvement of motor behavior,the proliferation of endogenous neural stem cells(ENSCs),the level of synaptophysin and the correlation of the number of ENSCs and the level of synaptophysin in the SOD1G93A GlH transgenic mouse model of ALS after FGF-2 administration.Methods;In this study,Rotarod test was used to estimate the motor behaviour,immunohistochemistry was used to marked the ENSCs and synaptophysin,the confocal counting system was used to estimate the number of ENSCs and synaptophysin in the SOD1G93A G1H mouse model before and after FGF-2 administration.The correlation of number of ENSCs and the level of synaptophysin was analyzed using Spearman statistical method.Results:Compared with ALS group,FGF-2 group showed a statistically significant improvement in motor behaviour,significant increase in the number of ENSCs and the level of synaptophysin.Conclusion:The neuro-protection mechanism of FGF-2 may associate with the proliferation of ENSCs and the level of synaptophysin,the study also provides foundation to the clinical therapy of ALS in the near future.The correlation of the number of ENSCs and the level of synaptophysin was analyzed positive.
