1 Objective 1.Through the herbalogical study and the "geographical distribution types of traditional Chinese medicine and high quality, authentic ingredients preferred mode",to study on genuine characteristics of Chuzhou Stemonae Radix. 2.To establish the exclusive quality evaluation of genuine medical herb Chuzhou Stemonae Radix which accomplished the excellent quality assessment of Chuzhou Stemonae Radix.2 Methods 1.Through the herbalogical study, the name, breeds, place of origin of Chuzhou Stemonae Radix was discussed in this paper, it confirmed that Chuzhou Stemonae Radix was the high-quality varieties after predecessors’ choice. 2.Study on genuine characteristics of Chuzhou Stemonae Radix based on the "geographical distribution types of traditional Chinese medicine and high quality, authentic ingredients preferred mode". 3.Study the quality evaluation system of Stemona sessilifolia(Miq.) Miq.,Stemona japonica(Bl.) Miq and Stemona tuberosa Lour.,establish microscopic identification,TLC,HPLC,HPLC-fingerprint of Chuzhou Stemonae Radix.Finally,we established quality standards of Chuzhou Stemonae Radix to present its high-quality and geoherbs. 4.With comparing mouse antitussive experiment of the different producing areas of Stemona sessilifolia(Miq.) Miq.,to confirm the antitussive of Chuzhou Stemonae Radix is better than other producing areas.3 Results 1. With herbalogical study, it confirmed that Chuzhou Stemonae Radix was the high-quality varieties after predecessors’ choice.Commentaries on the Illustrations was the relative complete national pharmacopoeia which was promulgated by the government.It recorded Chuzhou Stemonae Radix with figure and it proved Chuzhou Stemonae Radix was a widely used high-quality varieties. 2.From the geographical distribution types of Stemonae Radix,Stemona sessilifolia(Miq.) Miq. distributs in northern high latitudes.According to the endpoint hypothesis theory and geoherbs distribution law,Chuzhou Stemonae Radix was the high-quality varieties and geoherbs of Stemonae Radix. 3.There were significant difference of microscopic features of transverse section of root tuber of Stemona sessilifolia(Miq.) Miq.,Stemona japonica(Bl.) Miq and Stemona tuberosa Lour. From the thin layer chromatography of different producing areas of Stemona sessilifolia(Miq.) Miq.,Chuzhou Stemonae Radix had a unique geographical spot.Meanwhile,Chuzhou Stemonae Radix has a characteristic peak which was corresponding to the unique geographical spot in its HPTLC-fingerprint. The UPLC-QTOF-MS was used to identify unique geographical components of roots of Chuzhou Stemonae Radix.The component was possible Sessilistemonamine D. And established HPLC,HPLC-fingerprint of Chuzhou Stemonae Radix. 4.With comparing mouse antitussive experiment of the different producing areas of Stemona sessilifolia(Miq.) Miq.,the antitussive of Chuzhou Stemonae Radix is better than the producing areas of Henan under the same dosage.The result of statistical analysis showed that there were significant difference.4 Conclusion The facts mentioned above illuminate that Chuzhou Stemonae Radix had no obvious difference with Stemona japonica(Bl.) Miq and Stemona tuberosa Lour. in microscopic characteristics.Meanwhile,it had established the thin layer chromatography(TLC), HPTLC,HPLC,HPLC-fingerprint of Chuzhou Stemonae Radix,identify unique geographical components of Chuzhou Stemonae Radix and did the pharmacodynamics experiment.The established method is simple, specificity, reproducible, accurate and reliable.It can be used as an important basis for quality evaluation of Chuzhou Stemonae Radix.
