| Objective: The purpose of the study was to observe the effects of XiangLianHuaZhuoJieDu Decoction for Ulcerative Colitis model rats on general congditions,disease activity index score,pathological histology and IFN- γ and ERK mRNA of MAPK/ERK signal pathway,to explore the mechanism for the treatment of UC rats,and provided theoretical and experimental basis in the clinical application.Methods: 60 male Wistar rats weighing200g-250 g were randomly divided into six groups: the normal group(NG), the model group(MG), the low-dose XL group(LDG), the medium-dose XL group(MDG), the high-dose XL group(HDG), the SASP group(SASP), and each group has 10 rats. All groups except NG were controlled in order to make UC rats model by TNBS/ethanol. 3 days after the building, the NG continued to feed, the MG were given normal saline, the other groups received liquid gavage once a day lasted 2 weeks. Observed and recorded the weight and other general conditions,calculated DAI score,Sacrificed and dissected the rats and observed the colonic mucosa general concept, histopathological changes and score the histological disease. Detected the expression of IFN-γand MAPK/ERK by RT-PCR method.Results:1 The general conditions and DAI score of ratsCompared with NG,the rats in the other five groups both occurred lethargy,disordered lusterless hair,activity decrease and inflexible,their foodintake and weight declined,loose or mucobloody stool.After two weeks treatment,the general conditions of HDG were similar with NG,the improve in MDG and SASP werse no obvious compared with HDG,and the two groupswere no apparent difference,the improve in LDG werse worse than MDG and SASP,but better than MG.DAI score: There was statistically significant difference(P<0.05)between MG(2.50±0.47)with HDG(0.37±0.35)、MDG(0.90±0.32)、LDG( 1.50±0.62)、 SASP( 0.63±0.35); There was statistically significant difference(P<0.05) between HDG and MDG and LDG; There was no statistically significant difference(P>0.05) between HDL and SASP; There was no statistically significant difference(P>0.05) between MDG and SASP;There was statistically significant difference(P<0.05) between LDG and MDG、SASP.2 Histopathological observation of colonic mucosa and tissue and Histological disease scoreColonic mucosa: The MG was the worst. The HDG was closely to the NG. There was no significant difference between the MDG and the SASP,which were worse than the HDG, but better than the LDG.Pathological observation:Compared with NG, there was a small amount of colonic mucosal epithelial cell in MG,with loss gland and congested capillaries,obvious edema,and lots of inflammatory cells immersed, and development of corrosion or ulcer.Recoveried colonic mucosa in HDG was similarly to NG,which in MDG and SASP was worse than HDG but better than LDG.Histological disease score: The score of NG(0.21±0.16), was lower than the other five groups obviously(P<0.05);MG(3.25±0.40)was higher than every threatment group, and there was statistically significant difference between them(P<0.05); HDG(1.15±0.22) was lower than SASP(1.52±0.34) 、 MDG(1.55±0.29) and LDG(2.92±0.25), and there was statistically significant difference between them(P<0.05);SASP was lower than MDG, and there was no statistically significant difference between them(P>0.05); MDG was lower than LDG, and there was statistically significant difference between them(P<0.05).3 The expression of IFN-γ and ERK mRNAIFN-γ mRNA: The expression of IFN-γ mRNA in MG(0.80±0.13)was higher than in NG(0.23±0.04), and there was statistically significant difference between them(P<0.05); The expression of IFN- γ mRNA in all treatment groups was lower than MG, and there was statistically significant difference between them(P<0.05); The expression of IFN-γ mRNA in HDG(0.30±0.05)was lower than SASP(0.37±0.07)、MDG(0.46±0.05)、LDG( 0.57±0.06), and there was statistically significant difference between them(P<0.05); The expression of IFN- γ mRNA in MDG was lower than LDG, and there was statistically significant difference between them(P<0.05);The expression of IFN-γ mRNA in SASP was lower than MDG and LDG,and there was statistically significant difference between them(P<0.05).ERK mRNA: The expression of ERK mRNA in MG(0.27±0.02)was higher than in NG( 0.24±0.03), and there was statistically significant difference between them(P<0.05); the treatment groups was higer than MG,and there was statistically significant difference between them(P<0.05); The expression of ERK mRNA in HDG( 0.51±0.03) was higer than SASP(0.48±0.02), and MDG(0.47±0.02)and LDG(0.40±0.03), and there was statistically significant difference between them(P<0.05); The expression of ERK mRNA in SASP was higher than MDG, and there was no statistically significant difference between them(P>0.05); the MDG and SASP was higher than LDG, and there was statistically significant difference between them(P<0.05).Conclusions:1 XiangLianHuaZhuoJieDu Decoction can improve the diarrhea and blood in stool, alleviate the pathological injury and protect the gut mucosa of UC rats effectively. And there is a dose-effect relationship between its curative effect and dosage,while the effect of HDG is better than MDG and LDG obviously, and better than SASP.2 The mechanism of action of XiangLianHuaZhuoJieDu Decoction to treat UC is probably likely to down-regulating IFN- γ and activate MAPK/ERK signal pathway,to regulate the immune system, alleviateinflammation, promotes cell proliferation, inhibits apoptosis in colonic epithelial cells, and contribute to colon repair, so as to achieve the purpose of treatment of UC. |
