PART 1 Research on the Sustained Release Model Preparation of Cephalexin-PACA and the Drug Release Mechanism in Artificial Cerebraspinal FluidObjective: To research the Preparation of Cephalexin sustained release mode with PACA(poly cyano acrylic acid alkyl eater)materials as the carrier,and determine the release curve of Cefalexin sustained release model for artificial cerebrospinal fluid,and establish release model in vitro.Methods: This experiment adopts the gravity sink merhod for the preparation in artifical cerebrospinal fluid.The chromatographic condition for determination of Cep were C18 column and a mobile phase of water/ mathanol/3.86% acetic acid/ 4% sodium acetate(742:240:15:3,v/v),and the detection wavelength was set as 254 nm.Results: Cep in the sustained-release model could be sustained release for at least several weeks.Release curve in vitro was consistent with zero-order release,and the release speed and time were calculated.Conclusion: PACA materials can be used as slow release pipe,and can effectively realize that the effective components in PACA materials release slowly.It can be used as release carriers for drugs,and provide a simple,effective,economical and practical model for the drug sustained release stably in a long-term.PART 2 Determination of Oxytetracycline in tablets and feed by Luminol-Ni(IV)Complex Chemiluminescence System coupled and Mechanism ResearchObjective: A new method for the determination of oxytetracycline(OTC)by flow injection chemiluminescence.Methods: A novel flow injection analysis-direct chemiluminescence method has been developed for determination of OTC based on the enhancing effect of OTC on the CL reaction of luminol with an Ni(IV)complex in alkaline solution.Results: Under optimum conditions,for OTC,the linear range of2.0×10-8~8.0×10-7 g/mL with a detection limit of 1.0×10-12 g/mL,and the relative standard deviation(n=11)was 2.0% for 1.0×10-7g/mL OTC.Conclusion: The proposed method was applied in the analysis of OTC in tablets and feed sample with satisfactory result.PART 3 Determination of Demethylchlortet racycline in feed by Luminol-Ni(IV)Complex Chemiluminescence System and Mechanism ResearchObjective: A new method for the determination of demethylchlortet racycline(DETC)by flow injection chemiluminescence.Methods: A novel flow injection analysis-direct chemiluminescence method has been developed for determination of DMTC based on the enhancing effect of DMTC on the chemiluminescence reaction of luminol with an Ni(IV)complex in alkaline solution.Results: Under optimum conditions,for DMTC,the linear range of9.0×10-8~ 8.0×10-6 g/mL with a detection limit of 1.0×10-11 g/mL.The relative standard deviation(n=11)was 2.4% for 1.0×10-7g/mL DMTC.Conclusion: This method has broad linear range,high sensitivity,and convenience and speediness,it therefore,can be applied in the analysis of DMTC in feed sample with satisfactory result.
