| Background: Colorectal cancer is one of the common malignant neoplasm in the our country.The latest data shows that the mortality rate is ranked third,and it has a tendency to increase.This cancer is a serious threat to our life and wealth.Colorectal cancer genesis and development is a multifactor precess.CD226 molecule in the human body involved in a variety of immune responses,and some types of cancer are related.CD226 gene SNP was found with a variety of diseases.In recent years,the pathogenesis between CD226 gene SNP and tumor have been discovered by domestic and foreign scholars.According to current reports,a significant correlation has been found between CD226 gene SNP and gastric cancer,cervical squamous cell carcinoma,non-small cell lung cancer,but there is no clear-documented in colorectal cancer.Objective: This research will use a case-control study to explore the relationship of CD226 rs727088A/G and rs763361C/T polymorphism with the risk of colorectal cancer in Hebei province and around city Shijiazhuang.Methods: Choose the cases which were diagnosed colorectal cancer and treated in the fourth hospital of HEBEI medical university between Feb.2014 and Feb.2016.Totally 162 cases of rectal cancer and 209 cases of colon Cancer were collected.And 199 random healthy people were selected for doing case-control study.5 ml of venous blood was collected on the object of the experimental conditions including age,gender,smoking,alcohol consumption,family history and other information.The proteinase-K digestion-saturated sodium chloride salting out method and Polymerase Chain Reaction-ligase detection Reaction(PCR-LDR)were used to handle obtaining samples and detect CD226 gene rs727088A/G and rs763361C/T two SNP loci alleles and genotype.Experiment data use SPSS Ver19.0 software package(IBM Company in New York,Chicago,Illinois,USA)for statistical analysis.Age differences between the case and control groups use the t test;Gender,smoking,alcohol consumption,family history and so on difference in rectal cancer、colon cancer group and health control group and the distribution of allele frequency and genotype frequency distribution were conducted by chi-square test respectively.Hardy-Weinberg equilibrium was performed by comparing the observed and expected genotype by Chi-square test.Haplotype frequencies using EH software and 2LD software(1.2 version,Rockefeller University,New York).The ratio of relative risk ratio(odds ratio,OR)and 95% confidence intervals(confidence interval,CI)by non-conditional Logistic regression calculation obtained.There is significant difference of the standard when P < 0.05.Results:1 Demographic characteristicThe comparison of gender,age,smoking history and drinking history have no statistical significance between the colorectal cancer group and control group(P>0.05).The difference of family history between the colorectal cancer group and control group prove that family history maybe increase risk for colorectal in Hebei province by Chi-square test.(P value were 0.015 and 0.022 respectively in colon and rectal cancer group)2 H-W equilibrium analysis of CD226 rs727088A/G locus and rs763361C/T locusThe distribution of the genotype frequencies of rs727088A/G and rs763361C/T two loci were consistent with Hardy-Weinberg equilibrium(P>0.05).3 CD226 gene rs727088A/G polymorphism and colorectal cancer risk correlation analysisThe frequency of rs727088A/G CD226 gene loci A in the control group is 78.4%(312/398);66.0%(214/324)and 67.0%(280/418)in colon cancer and rectal cancer group respectively.In the comparison of two heavy,no statistical significance was found between haplotype A and G in colorectal cancer and the control group(both P<0.001).This result showed rs727088A/G CD226 gene loci G could rise risk for colorectal in Shijiazhuang city.The frequency of CD226 allele genotype AA、AG and GG were 61.3%(122/199)、34.2%(68/199)and 4.5%(9/199)in the control group,while the frequency are 41.4%(67/162)、49.4%(80/162)、9.2%(15/162)in colon cancer group and 42.1%(88/209)、49.8%(104/209)、8.1%(17/209)in rectal cancer group,which were significant difference(P value respectively 0.001,<0.001).CD226 rs727088 SNP have correlation with the incidence of colon and rectal cancer.4 rs763361C/T CD226 gene C/T polymorphism and colorectal cancer risk correlation analysisThe frequency of rs763361G/T CD226 gene loci T in the control group is 69.1%(305/398);66.0%(214/324)and 73.0%(305/418)in colon cancer and rectal cancer group respectively.Compared between each two groups respectively,P value are 0.384 and 0.223 respectively in colon and rectal cancer group,which show that in contrast between control group and the colon cancer group,there was no statistically significant.The frequency of CD226 rs763361C/T allele genotype CC、CT and TT are 46.7%(93/199)、44.7%(89/199)and 8.6%(17/199)in the control group,while GG、GT and TT genotype frequency are 43.8%(71/162)、44.4%(72/162)、11.8%(19/162)in colon cancer group and 53.1%(111/209)、39.7%(83/209)、7.2%(15/209)in rectal cancer group.In pairwise comparison between case and control group,the P value is 0.584 and 0.432,respectively in colon and rectal cancer group.Through the analysis of the results,carrying the G/T gene type of individual has no obviously relationship with colon cancer in Hebei province.Conclusion:1 CD226 gene rs727088A/G SNP may be correlated with the incidence of colorectal cancer risk,as well as,rs727088A/G CD226 gene loci G and AG,GG could rise risk for colorectal in Shijiazhuang city and the place around.2 CD226 gene rs763361C/T SNP may have no significantly correlation with colorectal cancer around city Shijiazhuang. |
