MicroRNAs is made up of 21-23 nucleotide highly conservative non-coding RNA,binding sites are usually SD sequence. Mi R- 218 is a tumor inhibitors had been reported many times, there is no report confirm the relationship between the microRNA and SUMO modification, but previous research has shown that microRNA is associated with many tumors. Ubiquitin is one of the important physiological processes in the human body, are necessary to human physiological and pathological reaction physiological reaction. Kind of ubiquitin protease 1 is by SENP1 gene encoding of a kind of ubiquitin enzyme, it helps to type 1, type 2, type 3 mature and make them out of protein. But now, on the function of these proteases, regulation and control as well as the specific research there are still many mysteries.Around the mi R- 218, this study mainly research on the role of SENP1 gene miR- 218, in turn, affects PC-3 cells proliferation, get the following conclusion:1. Mi R- 218 and selected genes SENP1 a certain amount of base pairing, in prostate cancer cells at the same time the high level expression of miR- 218. In prostate cancer cells at the same time the high level expression of mi R- 218.Using reverse transcription polymerase chain reaction(PCR) obtain recombinant plasmid miR- 218 and miR- NC. By plasmid PCR,double enzyme digestion show that sequencing appraisal right.2. By experimental verification, miR- 218 is able to control gene SENP1. MiR- 218 can go through and type of enzyme SENP1 gene 3 'UTR region combining SENP1 gene expression, and is verified by qPCR miR- 218 has a cut on BMI, LAMB3 and SENP1 gene.3. Through the experiment found that mi R- 218 to the class of the ubiquitin-proteasome pathway related proteins in expressing quantity has lower effect. SENP1 gene is an important factor to the SUMO change way, miR- 218 has inhibitory effect on gene SENP1, therefore,about the way the SUMO china-germany related proteins have inhibition.This study found that miR- 218 in prostate cancer cell PC-3 china-germany expression quantity highest, miR- 218 can cut SENP1 in PC-3 cells express, and affect the type of ubiquitin related protein expression in this way, thus inhibiting the activity of the prostate cancer cell PC-3, provide reliable experimental basis for follow-up study.
