Background Rheumatic heart disease,as a part of group A beta hemolytic streptococcus infectionis,caused by allergic reaction,which its pathogenesis is not clear.A majority of studies show that the pathogenesis of this disease is related to autoimmune and genetic factors,a minority of scholars believe that the pathogenesis is caused by inflammatory body NLRP-3 access number of conduction,but related research is less.Objective To investigate the role of NOD like receptor-protein-3(NLRP-3)inflammatory components(NLRP-3,apoptosis-associated speck-like protein containing CARD,ASC and caspase-1)in rheumatic heart valve calcification.Methods We screened the patients with heart disease and prolapse of mitral valve by cardiac surgery,which comes from the first affiliated hospital of Xin Xiang medical university between January 2013 and April 2014,and collected their mitral valves.Then divided the cases into control group(10 cases of mitral valve prolapse in patients with mitral valve)and the experimental group(50 cases of rheumatic heart disease mitral valve)according to the diagnosis criterion of rheumatic heart disease;We used the HE staining to show morphology of mitral valves;quantified calcium concentration by determination of calcium;and detected the expression of NLRP-3 inflammatory component by Western blotting.Results1.Compared with non-rheumatic heart disease mitral valve,HE staining showed that cell nuclears is pyknosising and disappearing light stenosis,moderate stenosis,severe stenosis of mitral valve in rheumatic heart disease;2.Compared with non-rheumatic heart disease mitral valve,The alizarin red staining results and determination of calcium ion showed that calcification area increasing in the mitral valve of rheumatic heart disease;3.Compared with non-rheumatic heart disease mitral valve,hen Western blot 、detected that the expression of(NLRP-3,ASC,caspase 1 and IL-1β)is increased in the mitral valve of rheumatic heart disease.Conclusion Rheumatic heart disease mitral valve lesion is related to NLRP-3 inflammatory component such as NLRP-3,ASC and caspase-1,and their expression up-regulation may increase the damage.
