Objective:We used different concentration of icariin(ICA) to act on human ovarian cancer SKOV3 cells in order to observe the effect of invasion and metastasis. Its potential molecular mechanism was discussed. Methods:1. The cells were divided into five groups: 0μg/mL ICA, 25μg/mL ICA, 50μg/mL ICA, 100μg/mL ICA, 200μg/mL of ICA.2. MTT assay was used to detect icariin’s impact on SKOV3 cell proliferation.3. Scratch wound assay was used to detect icariin’s impact on SKOV3 cell migration.4. Transwell chamber assay was used to detect icariin’s impact on SKOV3 cell invasive ability.5. ELISA kit was used to measure the expression of MMP-2 and MMP-9 protein of SKOV3 cell.6. Real Time-PCR was used to evaluate the expression of MMP-2 and MMP-9 mRNA of SKOV3 cell. Results:1. MTT assay showed that the SKOV3 cells were inhibited by ICA, and significant difference was showed between drug intervened groups and control group(P < 0.05).Moreover, with the increase of drug concentration,the inhibition of cells proliferation was significantly in does-dependent mariner in 100μg/mL concentration range.2. Scratch wound assay showed that the cell healing area of drug intervened groups(25μg/mL,50μg/mL, 100μg/mL) were statistically declined compared to the control group(P<0.05), which indicated that the ICA inhibited migration of SKOV3 cells.The inhibitory was significantly in does-dependent manner.3. Transwell chamber assay showed that the the invading cells transferred from upper surfaces to lower surfaces of drug intervened groups(25μg/mL,50μg/mL, 100μg/mL)were statistically declined compared to the control group(P<0.05), which indicated that the ICA inhibited invasion of SKOV3 cells. The inhibitory was significantly in does-dependent manner.4. ELISA kit revealed that the expression of MMP-2 and MMP-9 protein of drug intervened groups(25μg/mL, 50μg/mL, 100μg/mL) were statistically declined compared to the control group(P<0.05).The difference was statistically significant,and also has a dose-dependent manner.5. Reverse transcription polymerase chain reaction revealed that the expression of MMP-2 and MMP-9 mRNA of drug intervened groups(25μg/mL, 50μg/mL, 100μg/mL) were statistically declined compared to the control group(P<0.05). The difference was statistically significant,and also has a dose-dependent manner. Conclusions:All the results demonstrated that icariin plays an important role in the anti-metastasis and anti-invasion of ovarian cancer possibly by inhibiting the activity of MMP-2 and MMP-9.
