Objective: To compare the microscopic characteristics, TLC and HPLC of Moutan Cortex from different origins or different growth years, and to investigate the correlation between inorganic elements and its cultivating soil, the study tried to establish a quality evaluation system and quality standards for Moutan Cortex.Methods: The microscopic identification, TLC and content determination were used to evaluate the quality of Moutan Cortex from different origins or different growth years comprehensively refer to the Chinese pharmacopeia 2015. The two active ingredient(paeonol and paeoniflorin) were simultaneously detected by changing the extraction method and TLC plate, then established a new qualiatative identificaion for Moutan Cortex. On the basis of the TLC, the subject used HPLC dual-wavelength to detect the content of paeonol and paeoniflorin simultaneously after the extraction of paeonol and paeoniflorin by ultrasonic extraction and intended to develop quality standards of Moutan Cortex. The microwave digestion method was employed to process the medicinal materials and their planting soil, the suitable inorganic elements were choosed due to harmful element, routine index element principle and so on, then the inorganic elements were detected by ICP-MS technology. We adopt the similarity calculation software, SPSS, SIMCA-P and other data processing software for the analysis of the above results, thus the correlation between Moutan Cortex and its planting soil was researched.Results: There were significant differences in the level of Paeonol content in Moutan Cortex from different years and the same origin,the TLC plate spot color of two-year seedings was generally weak and the HPLC result showed that the content can not reach the requirement of the Chinese pharmacopeia 2015, the TLC spots of four years were the darkest and largest that contained the highest paeonol level, the level of paeonol started to decline from the beginning of the fifth year. Meanwhile, the study showed that the unshaved moutan was more than shaved moutan based on the same growth years and origins. The ICP-MS results showed that the enrichment of inorganic elements and the cultivating soil were correlated in some way after thecomparative analysis of Cortex Moutan from different origins or different growth years. Cu, Li, K, Na, Zn and so on showed a positive correlation, Cr, Hg and some other elements showed a negative correlation. On the same time, inorganic elements were not the same from differet growth years and the same origin.Conclusion: The quality evaluation method was established by the comparative analysis of Moutan Cortex from different origins or different growth years. On the basis of the primary part of the Chinese pharmacopeia 2015, the study tried to add the qualitative and quantitative analysis of Paeoniflorin which is another main active ingredient of Moutan Cortex, and perfected the the quality standard at the same time.The correlation between Moutan Cortex and its cultivating soil was researched, the quality evaluation and quality standard were supplemented based on the above research.
