| Objectives: This study was aim to study the protective effect of the shikonin in rats of acute pancreatitis complicated by acute lung injury established.Methods:A totally 24 healthy and clean male SD rats were randomly divided into the control group(C)(n=8),the acute pancreatitis group(A)(n=8)and the shikonin intervention group(I)(n=8).Experimental model of acute pancreatitis was made by retrograde injection of 4% sodium taurocholate(1ml/kg,0.2 mL/min)into the biliary-pancreatic duct.The shikonin intervention group were injected shikonin(50mg/kg,dissolved in DMSO)intraperitoneally at 30 minutes before making modes.Then to measure the level of serum Tumor Necrosis Factor α(TNF-α)and interleukin-6(IL-6),the expressions of phosphoinositide 3-kinase(PI3K),p-PI3 K and the activity of NF-κB in lung and the evaluation of pancreas pathology was according to Schmidt’s method.Result: The level of serum TNF-α and IL-6 in the intervention group was lower than that in the acute pancreatitis group.The expression level of MCP-1 and MIP-2 in intervention group were lower.The expression of NF-κB and p-PI3 K in the shikonin intervention group was less than that in the acute pancreatitis group.Compared with the control group,the lung tissue pathological score of the shikonin intervention group and acute pancreatitis group had significant difference.Conclusion:This study suggested that shikonin played a protective role in acute pancreatitis complicated by acute lung injury of rats.The protective mechanism may be related to the inhibition of PI3K-AKt-NF-κB passageway,so as to control the inflammatory reaction. |
